To observe the effects of Guben Jiannao Liquid(GBJNL) on learning and memory,sleep duration,number of behaviors and sleep-wake phase in Alzheimer′s Disease(AD) model rats,to explore the possible regulatory mechanism of GBJNL.Methods:The experimental rats were randomly divided into blank group,model group,GBJNL group and melatonin(MT) group,with 10 in each group.HomeCageScan 3.0 was used to detect and analyze the behaviors of the modeled rats in the cage for 24 h.Morris water maze was used to test the learning and spatial memory ability of rats in each group.The electroencephalography and electromyography were performed for 24 h using Sleepsign 3.0.Results:Morris water maze showed that compared with the conditions in the blank group,the escape latency of rats in the model group was prolonged,and the percentage of swimming time in target quadrant was decreased(P<0.05).Compared with the model group,the GBJNL group and the MT group had shortened escape latency and increased percentage of swimming time in target quadrant(P<0.05).HomeCageScan monitoring revealed that the model group had higher number of awakenings with reversed circadian rhythm,and lower sleep duration than the blank group(P<0.05),and after intervention,the number of awakenings in the GBJNL group was reduced with normal circadian rhythms,and the sleep duration was prolonged.The electroencephalography and electromyography by Sleepsign indicated that compared with the blank group,the model group had decreased accumulation of rapid eye movement(REM) sleep(P<0.05) and non-rapid eye movement(NREM) sleep(P<0.05),and after intervention,the number of awakenings in the GBJNL group was lower while the accumulation of NREM sleep was higher than those in the blank group(P<0.05).Conclusion:The AD model rats have a certain degree of circadian rhythm disorders,and GBJNL can improve the circadian rhythm disorders in them,increase the sleep quality and promote the sleep rhythm back to normal.
