To explore the active components in the target organ-brain tissue of Ruyi Zhenbao tablet,a classic Tibetan medicine,after repeated administration,and analyze its possible action characteristics.Methods:Six male SD rats were divided into a control group and an observation group,and were continuously administered for 7 days.The rats were sacrificed one hour after the last administration,and the blood plasma and brain tissue were collected.The components entering blood were analyzed,and the potential components entering the brain were detected and verified.Results:Full MS/dd-MS/MS model was used to qualitatively analyze the molecular weight composition of the first-order spectra of the target peak and the molecular ion peak of the second-order spectra.According to the components entering blood as the potential components entering the brain,they were input into the included list for sample detection.Combined with the results of databases mzCloud and ChemSpider,the potential components entering brain were searched.According to the retention time,primary mass number and secondary ion mass spectrometry data,a total of 13 components entering brain were found,among which daidzein,cholic acid,alfalfa ketone,3'-O-methylviolanone and piperine have been verified by standard substances.Tryptophan,anethole,isocostic acid and adenosine have been compared for secondary mass spectral fragment information through the mzCloud database.Conclusion:The effective components of drugs such as Dalbergiae Odoriferae Lignum,Piperis Longi Fructus,Licorice paste,Cinnamomi cortex,Aucklandiae radix,Hibiscus abelmoschus seeds,Calculus bovis,Carthami flos in Ruyi Zhenbao tablet can enter the target organ brain tissue and exert their curative effects.
