To investigate the therapeutic effect of Rebi formula on mice with gouty arthritis(GA) and the inhibition effects on inflammation factors interleukin-1β(IL-1β),interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α).Methods:Thirty C57BL/6 mice were randomly divided into control group,model group,low-dose Rebi formula group,high-dose Rebi formula group and colchicine group.Except for the control group,mice in the other four groups were injected with 3% monosodium urate crystal suspension near the plantar joint to generate mouse model of GA.Six hours after modeling,mice in the control group and the model group were given distilled water by intragastric administration.The mice in the low-dose and high-dose Rebi formula groups were given Rebi formula extracts(17.82 g/kg,35.64 g/kg) by gavage,and mice in the colchicine group were given colchicine solution intraperitoneally(0.5 mg/kg).The changes in body weight,foot swelling and thermal pain threshold were observed before and after the treatment.Hematoxylin-eosin staining was used to evaluate the changes of inflammation in foot tissue,liver and kidney.The contents of IL-1β,IL-6 and TNF-α in foot tissue were determined by ELISA.Gene expression levels of IL-1β,IL-6 and TNF-α in foot tissues were detected by qRT-PCR.Results:Compared with model group,high-dose Rebi formula or colchicine treatment significantly reduced the swelling degree of the hind paw of GA mice(P<0.01),improved the heat pain tolerance(P<0.001),and reduced the inflammatory cell infiltration of joint tissue.The protein and mRNA expression levels of inflammatory factors IL-1β,IL-6 and TNF-α were significantly decreased in joint tissue(P<0.001).In addition,there was no significant difference in the therapeutic effect between high-dose Rebi formula group and colchicine group(P>0.05).Conclusion:Rebi formula can effectively alleviate the symptoms and inhibit joint inflammation in mice with GA,with no significant effect on liver and kidney function.
