To explore the mechanism of action of Mahuang Xixin Fuzi Decoction(MXFD) and its decomposed formula on allergic rhinitis(AR),to determine whether it exerts anti-allergic effects through the nerve pathway Neuromedin U(NMU)/Neuromedin U receptor 1(NMUR1) on type 2 innate lymphoid cells 2(ILC2s).Methods:C57BL/6J mice were divided into seven groups according to single,double,and triple herbal combinations.AR models were prepared using ovalbumin(OVA).After successful molding,mice in the treatment groups were treated with the different herbal combinations of MXFD for 14 days.The success of the model was evaluated by the allergic reaction behavioral scoring method; the content of nuclear nactor of activated T cells 2(NFAT2) in lung tissues was detected by immunofluorescence double-staining; and the expression level of NMUR1 gene in lung tissues was detected by reverse transcription real-time quantitative polymerase chain reaction(RT-qPCR).Results:The mice were successfully modeled,with increased expression levels of NFAT2 and NMUR1 genes in all groups(P<0.01).After treatment with Chinese medicine,the allergic reaction behavioral scores of the treated groups were reduced(P<0.01); the expression of NFAT2 was reduced(P<0.01),with the Mahuang group had the most significant inhibitory effect; except for the Xixin Fuzi group,the expression of NMUR1 gene was reduced(P<0.01) in all other groups,with the Mahuang Fuzi and Xixin groups had the most significant inhibitory effect.Conclusion:MXFD can inhibit allergic rhinitis by affecting the NMU/NMUR1 pathway.Mahuang alone has the best effect in inhibiting NFAT2,while the combination of Mahuang and Fuzi,Xixin alone have the best effect on NMUR1 expression.
