Nucleoside Components of Pinelliae Rhizoma Praeparatum from Different Regions and Pharmacological Effect on Cold Fluid Obstructing the Lung in Mouse Model

    目的:比较河南、河北和甘肃3个产地法半夏7种核苷类成分的含量,并观察其对寒饮蕴肺模型小鼠的作用。方法:应用高效液相色谱法(HPLC)测定3个产地法半夏中7种核苷类成分的含量。以OVA、Al(OH)3和冷水浴刺激建立小鼠寒饮蕴肺模型并将不同产地的法半夏应用于寒饮蕴肺模型小鼠中,用酚红排泄量、氨水引咳评价化痰和镇咳作用,苏木精-伊红(HE)染色观察肺和气管组织病理变化,F4/80免疫组织化学法染色检测巨噬细胞浸润,酶联免疫吸附试验法(ELISA)检测炎症介质肿瘤坏死因子α(TNF-α)和白细胞介素10(IL-10)水平。结果:不同产地法半夏核苷含量有较大差异,河南半夏的尿嘧啶和腺苷的含量高(P<0.001),河北半夏中胞嘧啶核苷、尿嘧啶核苷和鸟嘌呤核苷的含量较高(P<0.001),甘肃半夏中腺嘌呤和β-胸苷含量较高(P<0.001)。在法半夏处理的寒饮蕴肺模型小鼠中,气管酚红排泄量增加,咳嗽次数降低(P<0.05)(酚红排泄量:河南组>河北组>甘肃组;咳嗽次数:河北组>甘肃组>河南组),支气管管腔病变明显减轻,气管组织增生和纤维化降低,肺和气管中巨噬细胞减少(P<0.05),血清中TNF-α水平降低(P<0.05),IL-10水平升高。结论:河南、河北和甘肃3个产地法半夏的核苷类成分含量有差别,3个产地法半夏均能减轻寒饮蕴肺模型小鼠中气道炎症反应水平和病理损伤,其中以河南产法半夏的祛痰镇咳效果最佳。

    Abstract:

    To compare the content of seven nucleoside components in Pinelliae Rhizoma Praeparatum from three different regions(Henan,Hebei,and Gansu),and observe their effects on a cold fluid obstructing the lung model in mice.Methods:High-performance liquid chromatography(HPLC) was used to determine the content of seven nucleoside components in Pinelliae Rhizoma Praeparatum from the three regions.A cold fluid obstructing the lung model in mice was established using OVA,Al(OH)3,and cold water bath stimulation.Pinelliae Rhizoma Praeparatum products from different regions were administered to the model mice,and the effects on phlegm resolving and cough relieving were assessed using phenol red excretion and ammonia-induced cough.Hematoxylin-eosin(HE) staining was performed to observe pathological changes in lungs and tracheal tissues.F4/80 immunohistochemistry staining was used to detect macrophage infiltration,and enzyme-linked immunosorbent assay(ELISA) was used to measure the levels of inflammatory mediators,including tumor necrosis factor-α(TNF-α) and interleukin-10(IL-10).Results:There were significant differences in nucleoside content among the Pinelliae Rhizoma Praeparatum products from the three regions.The content of uracil and adenosine was higher in the Henan Pinelliae Rhizoma Praeparatum(P<0.001),while cytidine,uridine,and guanosine were more abundant in Hebei Pinelliae Rhizoma Praeparatum(P<0.001).The Gansu Pinelliae Rhizoma Praeparatum had higher levels of adenine and β-thymidine(P<0.001).In the cold fluid obstructing the lung model,Pinelliae Rhizoma Praeparatum treatment increased phenol red excretion and decreased cough frequency(P<0.05),with the following pattern of effects:phenol red excretion(Henan>Hebei>Gansu) and cough frequency(Hebei>Gansu>Henan).The bronchial lumen damage was significantly alleviated,and the proliferation and fibrosis of tracheal tissues were reduced.The number of macrophages in lungs and tracheal tissues decreased(P<0.05),while serum TNF-α levels were lower(P<0.05),and IL-10 levels were higher.Conclusion:The nucleoside content in Pinelliae Rhizoma Praeparatum varies in Henan,Hebei,and Gansu.All three regional Pinelliae Rhizoma Praeparatum alleviated airway inflammation and pathological damage in the cold fluid obstructing the lung model mice.Among them,the Henan Pinelliae Rhizoma Praeparatum showed the best effect in dissolving phlegm and relieving cough.
