    摘要:

    目的:探讨酵母膏致高尿酸血症模型时是否同时伴有湿热内蕴证候表征,研究利清通颗粒的降血尿酸作用和清利湿热功效。方法:每天灌胃7.5 g/kg酵母膏造模,采用全自动生化仪检测血清尿酸(SUA)、肌酐(Scr)、尿素氮(BUN)、谷丙转氨酶(GPT)及谷草转氨酶(GOT)含量;酶联免疫吸附试验(ELISA)法检测血清中肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-2(IL-2)、白细胞介素-6(IL-6)、水通道蛋白3(AQP3)、水通道蛋白4(AQP4)、胃动素(MTL)、胃泌素(GAS)及醛固酮(ALD)含量;比色法检测血清中黄嘌呤氧化酶(XOD)、腺苷脱氨酶(ADA)含量;Western blot法检测肾脏组织中尿酸盐转运蛋白1(URAT1)、葡萄糖转运蛋白9(GLUT9)、三磷酸腺苷结合盒转运蛋白G2(ABCG2)、有机阴离子转运体1(OAT1),小肠组织中GLUT9、ABCG2的蛋白表达量。结果:与空白组比较,模型组血清SUA、GOT、Scr、MTL,ADA、肾URAT1,XOD、TNF-α、IL-1β、IL-6、ALD、肝脏XOD水平升高(P<0.05,P<0.01,P<0.001),肾ABCG2,小肠ABCG2,血清IL-2、GAS、AQP3、AQP4水平降低(P<0.05,P<0.01,P<0.001)。与模型组比较,利清通颗粒各剂量组血清SUA、ADA、肾组织URAT1,血清GOT,IL-1β、MTL含量降低(P<0.05,P<0.01,P<0.001);GAS含量升高(P<0.001)。低剂量组血清XOD,TNF-α、IL-6、ALD含量下降(P<0.05,P<0.01);AQP3含量升高(P<0.05)。高剂量组肝脏XOD、小肠组织GLUT9,血清XOD、TNF-α、ALD、IL-6含量下降(P<0.05,P<0.001);AQP4,IL-2、AQP3含量升高(P<0.01,P<0.001)。结论:酵母膏致高尿酸血症模型时同时伴有湿热内蕴证候表征,利清通颗粒具有降血尿酸作用和清利湿热功效。

    Abstract:

    To investigate whether the yeast extract-induced hyperuricemia model was accompanied by the syndromes of intrinsic dampness-heat and study the effect of Liqingtong Granules in lowering blood uric acid and clearing dampness-heat.Methods:The model was established by intragastric administration of 7.5 g/kg yeast extract daily.The contents of serum uric acid(SUA),creatinine(Scr),urea nitrogen(BUN),glutamic-pyruvic transaminase(GPT),and glutamic-oxaloacetic transaminase(GOT) were detected by the automatic biochemical analyzer.Serum levels of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),interleukin-2(IL-2),interleukin-6(IL-6),aquaporin 3(AQP3),aquaporin 4(AQP4),motilin(MTL),gastrin(GAS),and aldosterone(ALD) were detected by enzyme linked immunosorbent assay(ELISA).The contents of xanthine oxidase(XOD) and adenosine deaminase(ADA) in serum were determined by colorimetry.Western blot assay was used to detect the protein expression levels of urate transporter 1(URAT1),glucose transporter 9(GLUT9),adenosine triphosphate binding cassette transporter G2(ABCG2),and organic anion transporter 1(OAT1) in the kidney,as well as GLUT9 and ABCG2 in the small intestine.Results:Compared with those of the blank group,the levels of SUA,GOT,Scr,MTL,and ADA in the serum,as well as URAT1,XOD,TNF-α,IL-1β,IL-6,ALD in the kidney and XOD levels in the liver of the model group were increased(P<0.05,P<0.01,P<0.001).ABCG2 in the kidney,ABCG2 in the small intestine,and IL-2,GAS,AQP3,and AQP4 levels in the small intestine decreased(P<0.05,P<0.01,P<0.001).Compared with the model group,the contents of SUA and ADA in serum,URAT1 in the kidney,as well as GOT,IL-1β,and MTL in serum of all groups of Liqingtong Granules were decreased(P<0.05,P<0.01,P<0.001).GAS content increased(P<0.001).The contents of serum XOD,TNF-α,IL-6,and ALD in the low-dose group were decreased(P<0.05,P<0.01); AQP3 content increased(P<0.05).The contents of XOD in the liver,GLUT9 in the small intestine,and serum XOD,TNF-α,ALD,and IL-6 decreased in the high-dose group(P<0.05,P<0.001).The levels of AQP4,IL-2,and AQP3 increased(P<0.01,P<0.001).Conclusion:The yeast extract-induced hyperuricemia model is accompanied by the syndrome of intrinsic dampness-heat,and Liqingtong Granules have the effect of lowering blood uric acid and clearing dampness-heat.
