健脾益气摄血方对原发免疫性血小板减少症乏力小鼠线粒体功能的影响

    摘要:

    目的:探讨健脾益气摄血方治疗原发免疫性血小板减少症(ITP)乏力的作用机制。方法:将90只BALB/c小鼠按照血小板(PLT)计数分为正常组、模型组、泼尼松组及健脾益气摄血中剂量组、高剂量组,每组18只。比较各组PLT、小鼠负重游泳时间、琥珀酸脱氢酶复合物亚单位A(SDHA)、酪蛋白水解蛋白酶P(ClpP)、线粒体蛋白酶1(LonP1)蛋白和信使RNA(mRNA)表达。结果:与正常组比较,模型组小鼠PLT低,小鼠负重游泳时间短,SDHA、ClpP蛋白和mRNA表达高,LonP1蛋白和mRNA表达低(P<0.05);与模型组比较,健脾益气摄血方组PLT高,小鼠负重游泳时间长,SDHA、ClpP蛋白和mRNA表达低(P<0.05),健脾益气摄血中剂量组LonP1蛋白表达高(P<0.05),健脾益气摄血高剂量组LonP1mRNA表达较中剂量组高(P<0.05)。结论:健脾益气摄血方能有效提高ITP小鼠PLT,改善ITP乏力症状,其作用机制可能与调节线粒体关键蛋白酶SDHA、ClpP、LonP1表达,恢复线粒体功能有关。

    Abstract:

    To explore the mechanism of action of Jianpi Yiqi Shexue Formula in treating fatigue in primary immune thrombocytopenia(ITP).Methods:A total of 90 BALB/c mice were divided into the following groups based on platelet(PLT) count:normal group,model group,prednisone group,Jianpi Yiqi Shexue Formula medium-dose group,and high-dose group,with 18 mice in each group.The PLT,weighted swimming time,and protein and mRNA expression of succinate dehydrogenase complex subunit A(SDHA),caseinolytic protease P(ClpP),Lon protease 1(LonP1) were compared among the groups.Results:Compared with the normal group,the model group had lower PLT,shorter weighted swimming time,and higher SDHA and ClpP protein and mRNA expression,while LonP1 protein and mRNA expression was lower(P<0.05).Compared with the model group,the Jianpi Yiqi Shexue Formula group showed higher PLT,longer weighted swimming time,and lower SDHA and ClpP protein and mRNA expression(P<0.05).The medium-dose Jianpi Yiqi Shexue formula group exhibited higher LonP1 protein expression(P<0.05),while the high-dose group showed higher LonP1 mRNA expression than the medium-dose group(P<0.05).Conclusion:The Jianpi Yiqi Shexue Formula can effectively increase PLT in ITP mice and alleviate fatigue symptoms.Its mechanism may involve regulating the expression of key mitochondrial proteases SDHA,ClpP,and LonP1,thereby restoring mitochondrial function.
