    目的:探讨芪参还五胶囊(QSHWC)调节Janus激酶2(JAK2)/信号转导和转录激活子3(STAT3)通路对缺血性脑卒中(IS)大鼠神经元凋亡的影响。方法:SD大鼠分为IS组、对照组、QSHWC低剂量(QSHWC-L)组、QSHWC高剂量(QSHWC-H)组、丁苯酞组及QSHWC-H+库马霉素(JAK2激活剂)组。线栓法构建IS模型成功后立即给药处理。Zea-Longa评分法评价神经功能;分析大鼠踏空步数比例及脑梗死体积百分数;HE染色检测脑组织病理变化;酶联免疫吸附试验法(ELISA)检测肿瘤坏死因子-α(TNF-α)、细胞间黏附分子-1(ICAM-1)、白细胞介素-1β(IL-1β)水平;TUNEL染色法检测神经元凋亡;蛋白质免疫印迹法检测p53、p-JAK2、裂解胱天蛋白酶-3(Cleaved-caspase-3)、p-STAT3蛋白。结果:与对照组比较,IS组海马CA1区脑组织异常,神经功能评分、踏空步数比例、脑梗死体积百分数、神经元凋亡率、TNF-α、ICAM-1、IL-1β水平及p53、Cleaved-caspase-3、p-JAK2、p-STAT3蛋白表达均上调(P<0.05);与IS组比较,QSHWC-L组、QSHWC-H组、丁苯酞组海马CA1区脑组织病理损伤有所缓解,神经功能评分、踏空步数比例、脑梗死体积百分数、神经元凋亡率、TNF-α、ICAM-1、IL-1β水平及p53、Cleaved-caspase-3、p-JAK2、p-STAT3蛋白表达均下调(P<0.05);库马霉素逆转了高剂量QSHWC对IS大鼠炎症反应及神经元凋亡的抑制作用。结论:QSHWC可能通过抑制JAK2/STAT3通路抑制IS大鼠神经元凋亡。

    Abstract:

    To explore the effect of Qishen Huanwu Capsules(QSHWC) in regulating the Janus kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3) pathway on neuronal apoptosis in rats with ischemic stroke(IS).Methods:SD rats were divided into the IS group,control group,QSHWC low-dose(QSHWC-L) group,QSHWC high-dose(QSHWC-H) group,butyphthalide group,and QSHWC-H+Coumermycin(JAK2 activator) group.An IS model was established using the suture method,and treatment was administered immediately afterward.Neurological function was assessed using the Zea-Longa scoring method.The proportion of foot fault steps and the infarct volume percentage were analyzed.HE staining was used to examine brain tissue pathology.Enzyme-linked immunosorbent assay(ELISA) measured the levels of tumor necrosis factor-alpha(TNF-α),intercellular adhesion molecule-1(ICAM-1),and interleukin-1β(IL-1β).TUNEL staining was used to detect neuronal apoptosis.Western blot was employed to assess the expression of p53,p-JAK2,cleaved-caspase-3,and p-STAT3 proteins.Results:Compared with the control group,the IS group exhibited abnormal brain tissue in the hippocampal CA1 region,as well as elevated neurological dysfunction scores,proportion of foot fault steps,infarct volume percentage,neuronal apoptosis rate,TNF-α,ICAM-1,IL-1β levels,and expression of p53,cleaved-caspase-3,p-JAK2,and p-STAT3 proteins(P<0.05).Compared with the IS group,the QSHWC-L,QSHWC-H,and butyphthalide groups showed alleviated brain tissue damage in the hippocampal CA1 region,with reduced neurological dysfunction scores,proportion of foot fault steps,infarct volume percentage,neuronal apoptosis rate,TNF-α,ICAM-1,IL-1β levels,and expression of p53,cleaved-caspase-3,p-JAK2,and p-STAT3 proteins(P<0.05).Coumermycin reversed the inhibitory effects of high-dose QSHWC on inflammation and neuronal apoptosis in IS rats.Conclusion:QSHWC may inhibit neuronal apoptosis in IS rats by suppressing the JAK2/STAT3 pathway.
