To optimize the extraction process of Chinese medicine compound formulas using both central composite design-response surface methodology(CCD-RSM) and orthogonal design,in combination with the analytic hierarchy process-entropy weight method(AHP-EWM),and to provide a reference for selecting optimal Chinese medicine extraction methodologies.Methods:The comprehensive score,based on the transfer rates of puerarin,hesperidin,and liquiritin,as well as the extract yield,was used as the response value.Water-to-material ratio,extraction time,and number of extractions were selected as key process parameters.The AHP-EWM was applied to determine the weight coefficients of each evaluation index and calculate the comprehensive score.Both CCD-RSM and orthogonal design were used to optimize the extraction parameters,followed by experimental validation and comparison.Results:Using the extraction process of Shenling Chewable Tablets as a case study,the optimal extraction conditions obtained by the orthogonal design and the response surface methodology were different.However,in both methods,the relative standard deviations(RSD) of three validation batches were all below 3.0%,and the comprehensive scores closely matched the predicted values from each respective design.Conclusion:The response surface methodology offers more refined process optimization with good reproducibility and operability,making it more suitable than the orthogonal design for practical production of Shenling Chewable Tablets.
