    目的:建立肾虚血瘀型黄褐斑动物模型,研究鹿胎丹参白芍方的填精活血作用并探讨其作用机制。方法:将48只大鼠采用随机数字表法分为空白组、模型组、阳性药组及鹿胎丹参白芍方低、中、高剂量组,每组8只。除空白组,其他组大鼠给予紫外灯照射、肌内注射黄体酮、灌服羟基脲造模。从造模第1天开始,每天下午各组灌服受试药物。实验结束后,取照射部位皮肤和卵巢进行病理苏木精-伊红(HE)染色,测定各组酪氨酸酶(TYR)、内皮素(ET-1)、雌激素(E2)、卵泡刺激素(FSH)、抑制素B(INHB)、血栓素(TXB-2)等含量;并测定各组丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)、脂褐素(LPF)含量,用蛋白质印迹法检测皮肤中Kelch样环氧氯丙烷相关蛋白1(Keap1)、核因子E2相关因子(Nrf2)蛋白表达,探讨抗氧化祛斑作用机制。结果:与正常组比较,模型组皮肤可见褐色片状斑块;TYR、ET-1、INHB、TXB-2、MDA、LPF水平及Keap1蛋白表达升高(P<0.001),E2、FSH、CAT、GSH-Px含量及Nrf2蛋白降低(P<0.001)。与模型组比较,各给药组以上指标均得到改善。结论:通过灌服羟基脲、肌内注射黄体酮联合紫外线照射大鼠,可建立肾虚血瘀型黄褐斑模型;鹿胎丹参白芍方通过作用于Keap1/Nrf2抗氧化通路,抑制酪氨酸酶的氧化反应并提高卵泡质量,减少黑色素的形成,最终发挥中医填精活血功效及祛斑作用。

    Abstract:

    To establish an animal model of chloasma with kidney deficiency and blood stasis syndrome and to investigate the Lutai Danshen Baishao Formula's effects in replenishing essence and activating blood,and elucidate its underlying mechanisms.Methods:Forty-eight rats were randomly divided into six groups using a random number table:blank control,model,positive drug,and low-,medium-,and high-dose Lutai Danshen Baishao Formula groups(n=8 per group).Except for the blank group,rats were subjected to ultraviolet(UV) irradiation,intramuscular injection of progesterone,and oral administration of hydroxyurea to induce the model.From the first day of modeling,rats in the treatment groups received daily gavage of the respective test formulations.At the end of the experiment,skin and ovarian tissues from irradiated sites were collected for hematoxylin-eosin(HE) staining.The levels of tyrosinase(TYR),endothelin-1(ET-1),estradiol(E2),follicle-stimulating hormone(FSH),inhibin B(INHB),and thromboxane B2(TXB-2) were measured.Additionally,malondialdehyde(MDA),glutathione peroxidase(GSH-Px),catalase(CAT),and lipofuscin(LPF) levels were assessed.Western blot was used to detect the protein expression of Kelch-like ECH-associated protein 1(Keap1) and nuclear factor erythroid 2-related factor 2(Nrf2) in the skin to explore the antioxidant and anti-chloasma mechanisms.Results:Compared with the normal group,rats in the model group developed brown,patchy skin lesions.Levels of TYR,ET-1,INHB,TXB-2,MDA,LPF,and Keap1 protein expression were significantly increased(P<0.001),while E2,FSH,CAT,GSH-Px levels,and Nrf2 protein expression were significantly decreased(P<0.001).These abnormalities were ameliorated in all treatment groups.Conclusion:A rat model of chloasma with kidney deficiency and blood stasis syndrome can be successfully established by combining oral hydroxyurea,intramuscular progesterone,and UV irradiation.Lutai Danshen Baishao Formula exerts its therapeutic effects by modulating the Keap1/Nrf2 antioxidant pathway,inhibiting tyrosinase-mediated oxidation,and improving follicular quality,thereby reducing melanin formation and manifesting the traditional Chinese medicine effects of replenishing essence,activating blood,and eliminating chloasma.
