    目的:研究孕期应激诱导的先天肾虚体质仔鼠肺发育的影响及左归丸的调控作用机制。方法:40只雌性大鼠随机分为空白组、模型组、左归丸组(18.9 g/kg)及维生素E组(1.44 mg/kg)。除空白组外,其余组通过复合法刺激孕鼠直至分娩。检测糖水偏好试验、旷场实验及酶联免疫吸附试验法(ELISA)检测糖皮质激素(GC)、谷丙转氨酶(GPT)、谷草转氨酶(GOT)、血尿素氮(BUN)及肌酐(Cr)水平,苏木精-伊红染色(HE)孕鼠肝肾组织评估用药安全性;计算仔鼠只数,测量身长、体质量、脏器指数及苏木精-伊红染色观察肺组织发育情况;逆转录聚合酶链反应法(RT-PCR)及免疫荧光检测血管内皮生长因子受体的表达;蛋白质印迹法检测酪氨酸蛋白激酶2/信号转导与转录激活因子3通路(JAK2/STAT3)蛋白表达。结果:模型组孕期体质量、胎产数、糖水偏好、旷场实验显著降低,GC水平升高(P<0.05,P<0.01)。与模型组比较,左归丸组上述指标改善(P<0.05,P<0.01);各组孕鼠肝肾功能比较差异无统计学意义(P>0.05);模型组仔鼠体质量、胎产数、肺脏器比降低(P<0.05,P<0.01),与模型组比较,左归丸组上述指标升高(P<0.05,P<0.01)。左归丸组仔鼠肺组织形态改善。仔鼠出生当天,与模型组比较,左归丸组仔鼠肺组织VEGFR1、VEGFR2、VEGFR3 mRNA表达高(P<0.05),VEGFR2 mRNA表达高于VEGFR1、VEGFR3 mRNA表达(P<0.05),VEGFR2荧光信号高(P<0.01),JAK2、STAT3、p-JAK2、p-STAT3、VEGFR2蛋白表达高(P<0.05)。结论:左归丸能改善先天肾虚体质导致的子代大鼠肺发育异常,其作用机制可能与调节JAK2/STAT3信号通路有关。

    Abstract:

    To investigate the effect of prenatal stress-induced congenital kidney deficiency constitution on lung development in neonatal rats,and to explore the regulatory mechanism of Zuogui Pills in this process.Methods:Forty female rats were randomly divided into control,model,Zuogui Pills(18.9 g/kg),and vitamin E(1.44 mg/kg) groups.Except for the control group,all other groups were subjected to compound stress stimulation during pregnancy until delivery.Behavioral assessments included the sucrose preference test and open-field test.Enzyme-linked immunosorbent assay(ELISA) was used to measure serum levels of glucocorticoids(GC),glutamic-pyruvic transaminase(GPT),glutamic oxalacetic transaminase(GOT),blood urea nitrogen(BUN),and creatinine(Cr).Hematoxylin and eosin(HE) staining was performed on maternal liver and kidney tissues for safety assessment.The number of pups,body length,body weight,and organ indices of neonatal rats were recorded,and lung development was evaluated via HE staining.Reverse transcription-polymerase chain reaction(RT-PCR) and immunofluorescence were used to detect the expression of vascular endothelial growth factor receptors(VEGFRs).Western blot was employed to examine the expression of proteins in the Janus kinase 2/signal transducer and activator of transcription 3(JAK2/STAT3) signaling pathway.Results:Compared to the control group,the model group showed significantly reduced maternal body weight during pregnancy,lower litter size,decreased sucrose preference and open-field activity,and elevated GC levels(P<0.05,P<0.01).Zuogui Pills significantly improved these parameters compared to the model group(P<0.05,P<0.01).No significant differences were found in liver and kidney function among groups(P>0.05).Offspring in the model group exhibited reduced body weight,litter size,and lung organ index(P<0.05,P<0.01),all of which were improved in the Zuogui Pills group(P<0.05,P<0.01).Morphological improvement in lung tissue was observed in the Zuogui Pills group.On the day of birth,neonatal rats in the Zuogui Pills group showed significantly higher mRNA expression of VEGFR1,VEGFR2,and VEGFR3 compared to the model group(P<0.05),with VEGFR2 mRNA expression exceeding that of VEGFR1 and VEGFR3(P<0.05).VEGFR2 fluorescence signal and the expression levels of JAK2,STAT3,p-JAK2,p-STAT3,and VEGFR2 proteins were also significantly elevated(P<0.05).Conclusion:Zuogui Pills can improve abnormal lung development in offspring rats with congenital kidney deficiency constitution induced by prenatal stress.This effect may be mediated through regulation of the JAK2/STAT3 signaling pathway.
