    摘要:

    目的:建立黄芪新型颗粒饮片的质量评价标准和方法。方法:以传统的黄芪饮片为对照,通过评估流动性、出膏率、转移率、指标性成分含量、特征指纹图谱和相似度等指标,全面阐述新型颗粒饮片与传统饮片的品质差异。结果:DNA条形码鉴定表明不同批次黄芪饮片均为蒙古黄芪;流动性试验显示新型颗粒饮片以小粒径(24~65目)最佳,中粒径(10~24目)次之,传统饮片最差;出膏率结果显示新型颗粒饮片的大粒径(5~10目)最佳,中粒径次之,较传统饮片提升10%,小粒径最差,且小粒径在煎煮过程中出现糊化现象。指纹图谱分析表明新型颗粒饮片与传统饮片的化学成分相似度高(R>0.98);均一性分析显示新型颗粒饮片之间的化学成分相似度更高,一致性更好。指标性成分以毛蕊异黄酮葡萄糖苷含量计算,山西省忻州市五寨县产的中粒径和大粒径新型颗粒饮片显著高于原饮片(均P<0.05),以黄芪甲苷含量计算,3个产地的小粒径新型颗粒饮片均显著高于原饮片(均P<0.05)。结论:黄芪新型颗粒饮片与市售传统饮片的基本属性一致,但在流动性性、均一性方面表现更优,有利于临床调配和工业机械化生产,具有良好的市场潜力。

    Abstract:

    To establish the quality evaluation standard and method of novel Astragalus membranaceus granule decoction pieces.Methods:By employing the traditional Astragalus membranaceus decoction pieces as the control,the quality differences between the new granule decoction pieces and traditional decoction pieces were explained by assessing the indexes such as the flowability,paste rate,transfer rate,index component content,characteristic fingerprints,and similarity.Results:DNA barcoding identification revealed that various batches of Astragalus decoction pieces were all Mongolian Astragalus.The flowability test indicated that the novel granule decoction pieces had the best effect under small particle size(24 to 65 meshes),followed by the medium particle size(10 to 24 meshes),while the traditional decoction pieces exhibited the worst effect.The paste rate showed that the novel granule decoction pieces had the optimal effect under large particle size(5 to 10 meshes),followed by the medium particle size with a 10% improvement over the traditional decoction pieces.Additionally,the worst effect was observed under the small particle size.During the decoction process,the pieces with small particle sizes exhibited gelatinization.The fingerprint analysis showed that the chemical composition of the novel granule decoction pieces was highly similar to that of traditional decoction pieces(R>0.98).Homogeneity analysis revealed that there was higher similarity and consistency in the chemical composition of the novel granule decoction pieces.As the index component,the content of calycosin-7-O-β-D-glucoside in the pieces of medium and large particle sizes produced by five villages and counties in Xinzhou,Shanxi was significantly higher than that of the original decoction pieces(P<0.05).The content of astragaloside IV in the pieces of small particle size produced by three areas was significantly higher than that of the original decoction pieces(P<0.05).Conclusion:The fundamental characteristics of the new Astragalus membranaceus granules are in line with the traditional decoction pieces available in the market.However,the new pieces perform better in terms of flowability and homogeneity,conducive for clinical dispensing and industrial mechanized production with good market potential.
