To investigate the effects of angelica polysaccharide(AP) on rats with Hashimoto's thyroiditis(HT) based on the Wnt/β-catenin pathway.Methods:Of the 70 rats,ten were selected as the control group,with the remaining rats utilized to prepare HT rat models.The modeling rats were divided into the HT group,the group treated with different doses of AP(50 mg/kg,100 mg/kg,200 mg/kg),the group treated with positive drugs(Tripterygium wilfordii multiglucoside tablets),and the group treated with 200 mg/kg AP+Wnt/β-catenin signaling pathway activator-lithium chloride.The general condition of rats was observed.Thyroid function-anti-thyroglobulin antibody(TGAb),free thyroxine(FT4),anti-thyroid peroxidase antibody(TPOAb),thyroid stimulating hormone(TSH),and free triiodothyronine(FT3) were detected.The pathological damage of thyroid tissue was detected,and the mRNA expression of the cytokines interleukin(IL)-10,IL-17,IL-6,tumor necrosis factor(TNF)-α,apoptosis rate and the expression of Wnt/β-catenin signaling pathway proteins in the tissue were detected.Results:The levels of TPOAb,TGAb,TSH,mRNA expression of IL-17,IL-6,TNF-α,apoptosis rate,and the expression of Wnt1,β-catenin in serum of the HT group were significantly increased compared to the control group,with the levels of FT3 and FT4,and the expression of IL-10 mRNA significantly reduced(P<0.01).After treatment with AP and Tripterygium wilfordii multiglucoside tablets,the TPOAb,TGAb,TSH levels,mRNA expression of IL-17,IL-6,TNF-α,apoptosis rate,and protein expression of Wnt1,β-catenin were significantly reduced compared with the HT group,and the FT3 and FT4 levels and expression of IL-10 mRNA were notably increased,with the characteristic of dose dependence(P<0.05).Lithium chloride reversed the improvement effect of 200 mg/kg AP on thyroid function in rats with HT(P<0.01).Conclusion:AP can reduce the inflammatory response and apoptosis of rats with HT and improve thyroid function,which may be related to inhibiting the Wnt/β-catenin signaling pathway.
