    目的:探讨当归多糖(AP)基于Wnt/β-联蛋白(β-catenin)通路对桥本甲状腺炎(HT)大鼠的影响。方法:70只大鼠中选取10只为对照组,剩余进行HT大鼠模型制备,造模大鼠分为HT组、不同剂量当归多糖(50 mg/kg、100 mg/kg、200 mg/kg)组、阳性药物(雷公藤多苷片)组、200 mg/kg当归多糖+Wnt/β-catenin信号通路激活剂-氯化锂组;观察大鼠一般状况,检测甲状腺功能-抗甲状腺球蛋白抗体(TGAb)、游离甲状腺素(FT4)、抗甲状腺过氧化物酶抗体(TPOAb)、促甲状腺素(TSH)、游离三碘甲状腺原氨酸(FT3);检测甲状腺组织病理损伤,组织中细胞因子白细胞介素-10(IL-10)、IL-17、IL-6、肿瘤坏死因子-α(TNF-α)mRNA表达、细胞凋亡以及Wnt/β-catenin信号通路蛋白表达。结果:HT组血清中TPOAb、TGAb、TSH水平、IL-17、IL-6、TNF-α mRNA表达、细胞凋亡率、Wnt1、β-catenin表达较对照组显著增加,FT3、FT4水平、IL-10 mRNA表达显著降低(均P<0.01);经当归多糖及雷公藤多苷片治疗后,血清中TPOAb、TGAb、TSH水平、IL-17、IL-6、TNF-α mRNA表达、细胞凋亡率、Wnt1、β-catenin蛋白表达较HT组显著降低,FT3、FT4水平、IL-10 mRNA表达显著增加,具有剂量依赖性(均P<0.05);氯化锂逆转了200 mg/kg当归多糖对HT大鼠甲状腺功能的改善作用(P<0.01)。结论:当归多糖可以降低HT大鼠炎症反应及细胞凋亡,改善甲状腺功能,可能与抑制Wnt/β-catenin信号通路有关。

    Abstract:

    To investigate the effects of angelica polysaccharide(AP) on rats with Hashimoto's thyroiditis(HT) based on the Wnt/β-catenin pathway.Methods:Of the 70 rats,ten were selected as the control group,with the remaining rats utilized to prepare HT rat models.The modeling rats were divided into the HT group,the group treated with different doses of AP(50 mg/kg,100 mg/kg,200 mg/kg),the group treated with positive drugs(Tripterygium wilfordii multiglucoside tablets),and the group treated with 200 mg/kg AP+Wnt/β-catenin signaling pathway activator-lithium chloride.The general condition of rats was observed.Thyroid function-anti-thyroglobulin antibody(TGAb),free thyroxine(FT4),anti-thyroid peroxidase antibody(TPOAb),thyroid stimulating hormone(TSH),and free triiodothyronine(FT3) were detected.The pathological damage of thyroid tissue was detected,and the mRNA expression of the cytokines interleukin(IL)-10,IL-17,IL-6,tumor necrosis factor(TNF)-α,apoptosis rate and the expression of Wnt/β-catenin signaling pathway proteins in the tissue were detected.Results:The levels of TPOAb,TGAb,TSH,mRNA expression of IL-17,IL-6,TNF-α,apoptosis rate,and the expression of Wnt1,β-catenin in serum of the HT group were significantly increased compared to the control group,with the levels of FT3 and FT4,and the expression of IL-10 mRNA significantly reduced(P<0.01).After treatment with AP and Tripterygium wilfordii multiglucoside tablets,the TPOAb,TGAb,TSH levels,mRNA expression of IL-17,IL-6,TNF-α,apoptosis rate,and protein expression of Wnt1,β-catenin were significantly reduced compared with the HT group,and the FT3 and FT4 levels and expression of IL-10 mRNA were notably increased,with the characteristic of dose dependence(P<0.05).Lithium chloride reversed the improvement effect of 200 mg/kg AP on thyroid function in rats with HT(P<0.01).Conclusion:AP can reduce the inflammatory response and apoptosis of rats with HT and improve thyroid function,which may be related to inhibiting the Wnt/β-catenin signaling pathway.
