Abstract:To research the mechanism of Sanmiao Pill and Medicinal Cyathula root for inflammatory reaction of acute gouty arthritis in rats from the perspective of NF-κB signal conduction. Methods:Ten normal rats as the normal group, randomized other 60 rats of acute gouty arthritis to model group, Ermiao pills group, Sanmiao pill low dose group, Sanmiao pill high dose group, Cyathula low dose group and Cyathula high dose group. Right ankle joint was examined in each group. Measured the cell genes and NF-κB P65 protein expression level by ELISA assay immunohistochemistry and Western blot assay 3 days ofter intervention. Colored synovial tissue by HE staining and observed histopathological changes. Results: The medicine intervention groups' NF-κB P65 expression level in synovial tissue, the level of TNF-α IL-6 IL-8 and The total score of synovial tissue histopathology were significantly lower than that of the model group (P<0.01 or P<0.05). Conclusion: Sanmiao pill and Medicinal cyathula root maintained the level of cell genes TNF-α, IL-6, IL-8 by decreasing NF-κB P65 protein expression, and reduced the inflammatory reaction of acute gouty arthritis in the rats. Sanmiao pill high dose group was most effective, and Sanmiao pill was more effective than Ermiao pill. Medicinal cyathula root plays a role in targeting Ermiao pill at infected site.
