目的:观察养心通脉部位方(Active Principle Region of Yangxing Tongmai Formula,apr-YTF)对大鼠骨髓间充质干细胞(Bone Mesemchymal Stem Cells,BMSCs)移植心血瘀阻证心肌后分化为心肌样细胞的作用。方法:采用“冠脉结扎法”结合血流变学改变建立大鼠心血瘀阻证模型。然后将大鼠分为(apr-YTF)+BMSCs组、集落刺激因子(rhG-CSF)+BMSCs组、NS+BMSCs组、NS+DMEM组。原代培养大鼠BMSCs,体外溴氮胞苷(5-bromouracil Deoxyriboside Brdu)标记后注射至大鼠心肌,采用双染法观察心肌组织中心肌结蛋白Desmin-Brdu和肌球蛋白重链(Myosin Heavy Chain MHC)-Brdu的表达。结果:apr-YTF干预BMSCs移植心血瘀阻证心肌28 d后经心肌结蛋白Desmin-Brdu抗体染色后,NS+DMEM组梗死边缘未见双染表达,在NS+BMSCs组、(rhG-CSF)+BMSCs和(apr-YTF)+BMSCs组均可有猩红色Brdu抗体和棕色Desmin蛋白的同时表达,其中(rhG-CSF)+BMSCs和(apr-YTF)+BMSCs组阳性细胞计数高于NS+BMSCs组(P<0.05);(rhG-CSF)+BMSCs组和(apr-YTF)+BMSCs组两者之间差异无统计学意义(P>0.05)。结论:apr-YTF能促使移植于心血瘀阻证心肌环境后BMSCs向心肌细胞的方向分化。
Abstract:
To observe the effect of active principle region of site Yangxing Tongmai Formula(apr-YTF) improving the rat bone marrow mesenchymal stem cells (BMSCs) transplanted in myocardium with blood stasis syndrome differentiated into myocardial like cells.Methods:Coronary ligation combining blood rheology changes was used to set up rat blood stasis syndrome models.Then the rats were divided into (apr-YTF) + BMSCs group,recombinant human granulocyte colony-stimulating factor + BMSCs group,NS + BMSCs group,and NS + DMEM group.Primary cultured rat BMSCs,in vitro labeled by 5-romouracil deoxyriboside Brdu was injected into rat myocardium,using double staining myocardial tissue myocardial desmin Desmin-Brdu and myosin heavy chain (myosin heavy chain MHC)-Brdu expression.Results:After 28 days Desmin-Brdu myocardial desmin antibody staining,NS + DMEM group infarct border no double staining expressed in NS + BMSCs group,(rhG-CSF) + BMSCs and (apr-YTF) + BMSCs group can had scarlet and brown Desmin antibody Brdu simultaneous expression of proteins,including (rhG-CSF) + BMSCs and (apr-YTF) + BMSCs group-positive cell counts higher than NS + BMSCs group (P<0.05); (rhG-CSF) + BMSCs group and (apr-YTF) + BMSCs had no difference (P>0.05).Conclusion:Apr-YTF can promote BMSCs transplanted myocardial environmental blood stasis syndrome differentiation to cardiomyocytes .
