To compare the contents of 5-HMF between traditional decoction and granules of Shengmai San by HPLC.Methods:A HPLC method was applied on a Diamonsil C18 column(4.6 mm×250 mm,5 μm) by an elution using MeOH-H2O(10:90) as the mobile phase. The detection wavelength was at 280nm. The column temperature was set up at 30℃ and the flow rate was 1 mL/min.Results:The contents of 5-HMF in the traditional decoction from different comprise were from 7.56 to 19.48 mg/dose. The contents of 5-HMF in the granules from different comprise were from 7.26 to 23.22 mg/dose. Lilyturf and Schisandra chinensis both contain 5-HMF, granules of Shengmai San with no solubilizaion of 5-HMF, but by heating can increase the content of 5-HMF.Conclusion:The HPLC chromatotograms are similar in the two forms of Shengmai San, but the contents of 5-HMF in granules prepared from the same batches of medicinal materials is higher than that of the traditional decoction.
