      目的:从药用植物马甲子叶醇提物中分离筛选具有抗肿瘤作用的活性组分。方法:采用石油醚、乙酸乙酯、正丁醇萃取马甲子叶醇提物,并将这三个萃取部位进行体外抗肿瘤实验,再把筛选得到的具有较好抗肿瘤活性的部位进行硅胶柱层析分离,并采用MTT法对分离得到的组分进行体外抗肿瘤活性比较。结果:通过体外抗HepG-2细胞和HeLa细胞实验表明,乙酸乙酯萃取部位具有较好抗肿瘤活性。通过硅胶柱分离,获得5个组分,通过MTT实验发现这5个组分中,石油醚:乙酸乙酯为1∶1和1∶2洗脱所得到的两个组分具有明显的体外抗肿瘤活性,其中石油醚:乙酸乙酯为1∶2的洗脱具有更好的杀伤作用,对HepG-2细胞和HeLa细胞的IC50分别为45.79,13.16 μg/mL。结论:从马甲子叶醇提物乙酸乙酯部位中分离得到2种具有抗肿瘤作用的活性组分,为进一步分离抗肿瘤活性成分提供一定的指导作用。
English Summary:
      To separate and select anti-tumor activity part from the leaves of Paliurus ramosissimus. Methods: Extracts from the leaves of Paliurus ramosissimus were extracted with petroleum ether, ethyl acetate and n-butyl alcohol respectively. In vitro anti-tumor experiments were performed at the three extract sites. The optimal site with best anti-tumor activity then was screened by silica gel column chromatography, and MTT assay was used for the comparison of components obtained for antitumor activity. Results: In vitro anti HepG-2 cells and HeLa cell experiments showed that ethyl acetate fraction extracts had good anti-tumor activity. Separated by a silica gel column, five components were collected by MTT assay, which found that when the ratio of petroleum ether: ethyl acetate were 1∶1 and 1∶2, both eluted fraction group had in vitro anti-tumor activity and when the ration of the elution of petroleum ether to ethyl acetate was 1∶2 the anti-tumor function was better. IC50 of HepG-2 cells and HeLa cells were 45.79 and 13.16 μg·mL-1 respectively. Conclusion: Two kinds of active ingredients with anti-tumor effect were separated from the leaves of Paliurus ramosissimus by ethyl alcohol, which may provide guidance for the further separation of anti-tumor active ingredient.
