      目的:观察参元丹对缺血再灌注心肌细胞内质网应激相关因子Grp78、CHOP表达的影响。方法:离体培养新出生1~3 d大鼠心肌细胞,并分为正常组、模型组、参元丹组及空白血清组。应用三气培养箱制造缺血(2 h)/再灌注(4 h)模型。采用RT-PCR法测定内质网应激相关因子GRP-78、CHOP因子的表达。结果:模型组细胞内Grp78、CHOP表达均明显上升(P<0.01)。与模型组比较10%参元丹组中,Grp78(P<0.01)及CHOP(P<0.05)均有明显下降,10%空白血清组Grp78、CHOP较模型组无明显变化。结论:参元丹可以通过减轻内质网应激减少心肌细胞的凋亡,保护心肌细胞。
English Summary:
      To observe the effect of Shen Yuan Dan(SYD) on Grp78 and CHOP expression in ischemia-reperfusion myocardium cells. Methods: To culture cardiomyocytes from newborn rats, and then divided them into normal group, model group, SYD group and blank serum group. Ischemia (2 h) / reperfusion (4 h) model was established by tri-gas incubator. Real-time polymerase chain reaction (PCR) were used to detect the Grp78 and CHOP gene expression levels. Results: The Grp78, CHOP expression levels in model group were significantly increased (P<0.01). Compared with model group, Grp78 and CHOP were significantly decreased in 10% SYD group (P<0.05). Levels of Grp78 and CHOP has no obvious change between 10% blank serum group and model group. Conclusion: SYD can reduce endoplasmic reticulum stress in myocardial cell apoptosis and protect myocardial cells.
