      目的:观察TGF-β1、MMP-9在哮喘大鼠气道重建模型中的动态变化,探讨丹参干预气道重建的作用机制。方法:SPF级SD成年雄性大鼠70只随机分为正常对照组、哮喘模型组(哮喘2周组、哮喘4周组、哮喘8周组)、丹参干预组(丹参2周组、丹参4周组、丹参8周组)3个大组,7个小组。用卵蛋白(OVA)-氢氧化铝混悬液致敏与激发建立大鼠哮喘气道重塑模型,激发2、4、8周后处死。观察哮喘大鼠肺组织病理变化,采用ELISA法检测大鼠肺组织TGF-β1、MMP-9水平。结果:哮喘大鼠气道炎性细胞浸润及支气管平滑肌增厚,随着激发时间的延长逐渐加重;同时,哮喘大鼠肺组织TGF-β1、MMP-9水平逐渐升高,哮喘4周组、哮喘8周组较正常对照组显著升高(P<0.05、P<0.001);使用丹参干预后,丹参8周组较哮喘8周组显著降低(P<0.001)。结论:气道重建的程度随着哮喘发作及时间推移而发生动态变化;TGF-β1、MMP-9参与哮喘气道重建过程;活血化瘀药丹参可通过抑制TGF-β1、MMP-9水平而干预阻断哮喘气道重建。
English Summary:
      To observe the dynamic changes of TGF-β1 and MMP-9 in the airway remodeling of asthma rat model and explore the mechanism of salvia miltiorrhiza (Danshen) in intervening airway remodeling. Methods: Seventy healthy adult male rats were randomly divided into the normal control group, asthma model group (subdivided into two-week asthmatic group, four-week asthmatic group and eight-week asthmatic group) and salvia miltiorrhiza group (subdivided into two-week salvia miltiorrhiza group, four-week salvia miltiorrhiza group, and eight-week salvia miltiorrhiza group) with a total of seven groups with 10 rats in each. Asthmatic airway remodeling rats model was established with ovalbumin and Al(OH)3, and rats were observed 2, 4 and 8 weeks after induced model respectively. The pathological changes of the lung tissue were observed. The level of TGF-β1 and MMP-9 in lung tissue was detected by ELISA. Results: Airway inflammatory cells infiltrated in airway tissue and the thickness of bronchial smooth muscle increased. With the prolongation of the stimulating time, TGF-β1 and MMP-9 level increased gradually in the asthma model group. Compared with the normal control group, there were significant differences (P<0.05, P<0.001) in the four-week asthmatic group and eight-week asthmatic group. After salvia miltiorrhiza was applied, the TGF-β1 and MMP-9 level decreased. Compared with the eight-week asthmatic group, there were significant differences (P<0.001) in the eight-week salvia miltiorrhiza group. Conclusion: The changes of airway remodeling in asthma are dynamic process and TGF-β1and MMP-9 may intervene in the changes of airway remodeling process. Salvia miltiorrhiza improve the state of airway remodeling in asthmatic rats by inhibiting the level of TGF-β1 and MMP-9.
