目的:研究除幽愈疡免煎颗粒对乙酸致大鼠实验性胃溃疡的愈合效果。方法:将清洁级SD大鼠随即分为5组:A组(正常组/对照组)、B组(模型组)、C组(西药治疗组)、D组(中西医结合组)及E组(纯中药治疗组)。各组均10只,雌雄各半。采用冰乙酸腐蚀法制作胃溃疡大鼠模型。观察除幽愈疡免煎颗粒干预胃溃疡大鼠模型的胃黏膜病理形态学变化,酶联免疫(ELISA)方法检测血清标本VEGF含量变化。免疫组化法检测各组大鼠VEGF表达,Real-Time PCR检测各组大鼠VEGF表达。结果:同模型组相比,中药、西药和中西药组溃疡面积均显著缩小,差异均有统计学意义(P<0.05);同时分别与西药组、中西药组相比,中药组溃疡面积均显著缩小,差异都有统计学意义(P<0.05);中药组溃疡抑制率为62.8%,高于中西药组27.1%和西药组34.6%。免疫组织化学结果表明,正常组、中药组和中西医结合组大鼠胃溃疡黏膜有较强的VEGF蛋白表达,其余2组VEGF蛋白表达较弱;其中,对照组中VEGF mRNA表达水平显著高于其他组,而中药组和中西药结合组中VEGF mRNA表达水平与对照组相近,明显高于模型组与西药组,差异有统计学意义(P<0.05)。ELISA分析发现,中药组血清VEGF水平与对照组水平相当,而西药组、中西药结合组中VEGF水平低于其他组,差异有统计学意义(P<0.05)。结论:除幽愈疡免煎颗粒可增强胃溃疡大鼠胃组织VEGF及VEGFmRNA的表达,促进血管新生,促进溃疡愈合。

English Summary:

To assess the effect of Chuyouyuyangmianjian granule on healing process of acetic-acid-induced gastric ulcer in rats.Methods:The rat model of gastric ulcer was constructed by smearing acetic acid method.Speci?c pathogen-free SD mice were randomly classified into five groups：group A(normal control)，group B(acetic-acid-induced gastric ulcer)，group C(western medicine treatment)，group D(combined therapy of Chinese and Western medicine)，group E(western medicine treatment).Each group contained five female and five male rats and was matched by body weight and age.The protein and mRNA expression levels of vascular endothelial growth factor(VEGF)，in the gastric ulcer tissues or serum were examined by immunohistochemical analysis，ELISA and real-time PCR，respectively.Results:The ulcer areas in group C，D，E were significantly reduced compared with that in group B(P<0.05).Notably，the ulcer area in group E was significantly reduced compared with those in group C and group D(P<0.05).The inhibition rate of Chinese medicine was 62.8%，higher than those of combined therapy of Chinese and Western medicine(27.1%)and western medicine treatment(34.6%).The results of immunohistochemical analyses showed that the expression level of VEGF was upregulated in the gastric ulcer tissues of group A，D，E compared with those of the other groups.Using real-time PCR analyses，we found that the level of VEGF mRNA in group A was the highest among the levels of all groups，and the levels in group D and group E were significantly higher than those in group B and group C(P<0.05).In addition，there was no statistically significant difference of serous VEGF level between group A and group E through ELISA analyses.The level of VEGF in serum of group C and group D were lower than those of the other groups.Conclusion:Chuyouyuyangmianjian granule may improve the healing of gastric ulcer through upregulating the level of VEGF.
