      目的:观察加味良附丸联合5-FU对荷人胃癌裸鼠血清TGF-β1、IL-17水平的影响。方法:60只裸鼠造模成功后随机分为模型组、5-FU组、加味良附丸中剂量+5-FU组(联合给药组)及加味良附丸高、中、低剂量组。加味良附丸高、中、低剂量组给药剂量分别为10 g/(d·kg)、5 g/(d·kg)、2.5 g/(d·kg);5-FU组给药剂量为17 mg/(d·kg);模型组给予等体积生理盐水,联合给药组剂量为加味良附丸中剂量5 g/(d·kg)联合应用5-FU(17 mg/(d·kg)。连续给药10 d,眼球取血,ELISA法检测血清TGF-β1、IL-17含量。结果:联合给药组、5-FU组、加味良附丸高、中、低剂量组裸鼠血清中TGF-β1、IL-17含量明显低于模型组(P<0.01);联合给药组血清TGF-β1含量与新加良附高丸剂量组、5-FU组比较,差异无统计学意义(P>0.05),与加味良附丸中、小大剂量组比较,差异有统计学意义(P<0.05);各给药干预组血清IL-17含量差异无统计学意义(P>0.05)。结论:炎性反应细胞因子TGF-β1、IL-17可能参与了胃癌发生发展,加味良附丸联合5-FU具有抑制人胃癌细胞SGC-7901移植瘤裸鼠血清中炎性反应细胞因子TGF-β1、IL-17的表达,其可能通过调节炎性反应细胞因子的表达来影响胃癌的发生与发展。
English Summary:
      To observe the effects of Supplemented Liang-fu Pill(SLFP) combined with 5-FU on TGF-β1、IL-17 level in serum of SGC-7901 tumor bearing nude mice. Methods: The tumor-bearing nude mice models with human gastric cancer cell line SGC-7901were established and randomly divided into six groups: model group, 5-FU group, combination group and SLFP in high, medium, low dose groups. The dosage of SLFP in high, medium, low dose groups were 10 g·d-1·kg-1、5 g·d-1·kg-1、2.5 g·d-1·kg-1 respectively. The 5-FU group′s dosage is 17 mg·d-1·kg-1, the model group were given equal normal saline and the combination group were given Supplemented Liang-Fu Pill (5 g·d-1·kg-1) and 5-FU(17 mg·d-1·kg-1). The mice were treated for 10 days and then detected for the level of TGF-β1 and IL-17 in serum of SGC-7901 tumor bearing nude mice with the method of ELISA. Results: The levels of TGF-β1 and IL-17 in 5-FU group, combination group, SLFP in high, medium, low dose groups were significantly lower than those in model group (P<0.01). The levels of serum TGF-β1 in the combination group was not significantly different compared with that of SLFP in high dose group and the 5-FU group (P<0.05). But there was significant different compared with SLFP in high, medium and dose groups. There were no significant difference of the levels of serum IL-17 in each group(P>0.05). Conclusion: Inflammatory cytokine TGF-β1, IL-17 may result in gastric cancer. The SLFP combined with 5-FU maybe inhibit the expression of TGF-β1and IL-17 in the serum of human gastric cancer cell SGC-7901 tumor bearing nude mice, which may affect the growth of gastric cancer by regulating the expression of inflammatory cytokines.
