Abstract:To explore the protective effects of Uygur medicine Nigellae Semen on acute alcoholic liver damage mice and its inhibition on NF-кB signaling pathway.Methods:After establishing acute alcoholic liver injury mice model,50 mice were randomly divided into blank group,model group,positive control group,nigellae semen extraction high dose (6 g/kg) group,and nigellae semen extraction low dose (2 g/kg) group.Except for the blank group,the other groups were orally administered 56 degree liquor 10 mL/kg for 15 days.After 10hrs of food forbidden but no water forbidden,observe the general state of mice,detect their body weight,liver weight,liver index,liver function indexes of alanine aminotransferase (ALT),aspartate aminotransferase (AST),the expression of NF-кB and TNF-α in liver tissue and liver tissue pathological changes.Results:Compared with the blank group,the body weight and liver index of the model group decreased,and serum ALT and AST activity significantly increased (P<0.05).Compared with the model group,the liver weight of mice,body weight and liver index increased in nigellae semen groups,while serum ALT and AST activity decreased significantly (P<0.05).Model group mice showed significantly hepatocytes ballooning degeneration,necrosis and accompanied by a small amount of inflammatory cells infiltration.Compared with the model group,the nigellae semen group showed lower pathological changes.Compared with the blank group,the serum concentrations of NF-кB and serum TNF-α content significantly increased in model group.Compare with the model group,the serum concentrations of NF-кB and serum TNF-α content significantly decreased in the nigellae semen groups.Conclusion:Nigellae semen extraction has a protective effect on acute alcoholic liver damage mice,and it can inhibit the expression of NF-кB and TNF-α liver cells.
