目的:观察榄香烯乳联合人参皂苷方案对卵巢癌细胞耐药株A2780/PTX的影响作用。方法:采用体外培养的卵巢癌细胞耐药株A2780/PTX进行实验,随机分成4组,空白组:新鲜的全培养液;人参皂苷组:30 μg/mL人参皂苷培养液;榄香烯乳组:20 μg/mL榄香烯乳培养液;联合组:30 μg/mL人参皂苷+20 μg/mL榄香烯乳培养液。通过流式细胞仪检测细胞周期;通过MTT法检测榄香烯乳及人参皂苷单用的细胞毒作用及2者联合应用时的作用;采用Western-blot观察A2780/PTX细胞内蛋白的表达;采用激光共聚焦显微镜检测线粒体膜电位的变化。结果:流式细胞术检测结果显示,与人参皂苷组、榄香烯乳组及空白组比较,联合组在G0/G1期细胞有明显增加,差异有统计学意义(P<0.05),A2780/PTX细胞被阻止在G1期;MTT法检测结果显示不同浓度的药物剂量可以对卵巢癌细胞产生抑制作用,并呈一定的剂量依赖性;Western blotting结果显示,A2780/PTX细胞中联合组蛋白表达较空白组、人参皂苷组、榄香烯乳组显著降低(P<0.05);联合组A2780/PTX细胞株线粒体膜电位下降较人参皂苷组、榄香烯乳组及空白组更为明显(P<0.05)。结论:榄香烯乳联合人参皂苷方案能对卵巢癌细胞耐药株A2780/PTX产生明显的生长抑制作用。2者联合应用可以提高有效率,降低不良反应。

English Summary:

To observe the elemi olefinic emulsion combined ginseng saponin scheme on ovarian cancer cell A2780/PTX impact resistant strains.Methods:Using vitro cultured ovarian cancer cell A2780/resistant strains PTX experiment，randomly divided into 4 groups：blank group：just add fresh whole culture; Ginseng saponin groups：30 mu g/mL culture ginsenosides; Elemi ene group：20 mu g/mL olive incense ene milk culture medium; Joint group：30 mu g/mL + 20 mu g/mL ginsenosides elemi milk culture medium.By flow cytometry instrument to detect cell cycle; Lam incense ene milk by determined by MTT method and use the cytotoxic effect of ginsenosides and their joint application; Using Western blot observation A2780/PTX protein expression in the cell; With laser confocal microscope detection of mitochondrial membrane potential changes.Results:Flow cytometry detection，according to the results and ginseng saponin，lam incense ene milk group and the blank group comparison，the joint group of cells in G0/G1 phase increased markedly，and the statistical significant difference(P<0.05)，A2780/PTX cells in G1 phase is blocked; Determined by MTT method to detect the result shows that different concentrations of doses can inhibitory effect on ovarian cancer cells，and a certain dose dependent;，according to the results of Western blotting A2780/zhonglian PTX cells to form protein expression than blank group，ginseng saponin，lam incense ene group significantly reduced(P<0.05); Joint group A2780/PTX cell lines of mitochondrial membrane potential decreased the ginseng saponins group，lam incense ene group and blank group is more obvious(P<0.05).Conclusion:Lam incense ene milk can joint scheme ginsenosides on ovarian cancer cell A2780/PTX resistant strains produce obvious growth inhibition.The joint application can improve the efficient，reduce the adverse reaction.
