    摘要:

    目的:探讨复方益智颗粒对Aβ25-35导致PC12细胞损伤的保护作用,为进一步研究该产品改善阿尔茨海默症记忆功能障碍的作用机制奠定基础。方法:取经不同处理的PC12细胞分为正常对照组、Aβ25-35模型组、Aβ25-35+正常血清组、Aβ25-35+CYZG含药血清组(不同浓度)。正常对照组:DMEM培养基培养48 h,Aβ25-35模型组:DMEM培养基培养24 h后换含20 μM Aβ25-35的DMEM培养基继续培养24 h,Aβ25-35+正常血清组:正常血清预处理24 h后加入20 μM Aβ25-35的DMEM培养基继续培养24 h,Aβ25-35+CYZG含药血清组:不同浓度CYZG含药血清预处理24 h后,加入20 μM Aβ25-35的DMEM培养基继续培养24 h。分别采用MTT法检测细胞增殖情况、流式细胞仪检测PC12神经细胞凋亡情况。结果:Aβ25-35时间依赖性地抑制PC12细胞的生长,不同浓度CYZG含药血清对其造成的细胞损伤有保护作用,随着含药血清剂量的增加,细胞的存活率增加,生长抑制率减少。Aβ25-35能显著引起PC12细胞凋亡,不同浓度CYZG含药血清对其造成的细胞凋亡有抑制作用。结论:复方益智颗粒对Aβ25-35引起的PC12细胞损伤具有一定的保护作用,其神经保护作用可能与抑制Aβ25-35引起的PC12细胞凋亡有关。

    Abstract:

    To observe the protection effects of Fufang yizhi Granule on the cellular damage induced by the protein Aβ25-35,and to be the basis of further study on the mechanism of this product to improve the memory in Alzheimer’s disease. Methods: Different processing cells were divided into Normal control group, Aβ25-35model control group, Aβ25-35+Normal serum group, and Aβ25-35+CYZG serum group, then given different procedure. Normal control group: cultured with DMEM medium for 48 h, Aβ25-35model control group, 24 h after being cultured with DMEM medium, then added the medium contained 20 μMAβ25-35and continuously cultured 24 h. Aβ25-35+Normal serum group: 24 h after being cultured with DMEM medium contained normal serum, then add the medium contained 20 μMAβ25-35and continuously cultured 24 h, Aβ25-35+CYZG serum group: 24 h after being cultured with DMEM medium contained CYZG serum, then add the medium contained 20 μMAβ25-35and continuously cultured 24 h. Results: Protein Aβ25-35 could time-dependently inhibit the growth of PC12 cells, CYZG serum of different concentrations had protective effect on the cellular damage, with the increase of the CYZG serum dosage, the cell survival rate increase, and growth inhibition rate decrease. Protein Aβ25-35 could induce the apoptosis of PC12 cells, CYZG serum of different concentrations significantly inhibited the apoptosis. Conclusion: Fufang yizhi Granule could protect the PC12 from the damage induced by Protein Aβ25-35, the mechanism of neuroprotective effect of Fufang yizhi Granule may relate with the inhibition effect of apoptosis.
