      目的:观察脉抒(MaiShu,MS)对高脂血症金黄地鼠血脂和血管内皮功能的影响。方法:以高脂膳食诱导高脂血症金黄地鼠模型,给予不同剂量脉抒(低剂量:1 g/kg/d、中剂量:2 g/kg/d、高剂量:4 g/kg/d),观察给药2、4、8周后血清总胆固醇(Total Cholesterol,TC)、三酰甘油(Triglyceride,TG)、高密度脂蛋白胆固醇(High Density Lipoprotein Cholesterol,HDL-C)、非高密度脂蛋白胆固醇(non-high Density Lipoprotein Cholesterol,non-HDL-C)的变化;采用快速蛋白液相色谱(Fast Protein Liquid Chromatography,FPLC)技术分离给药8周后血清中极低密度脂蛋白(Very Low Density Lipoprotein,VLDL)、低密度脂蛋白(Low Density Lipoprotein,LDL)、高密度脂蛋白(High Density Lipoprotein,HDL),并检测其所含的总TC含量;测定肝脏TC、TG含量,计算肝指数;采用全自动血液黏度仪检测8周后全血血黏度值;采用离体微血管压力直径灌流系统观察各组金黄地鼠肠系膜小动脉对不同浓度梯度乙酰胆碱(acetylcholine,Ach)诱导的内皮依赖性舒张反应的变化,评价脉抒对血管内皮功能的影响。结果:脉抒高、中剂量在2、4、8周均能明显降低高脂血症金黄地鼠血清TC含量(P<0.01、P<0.01、P<0.05;P<0.01、P<0.01、P<0.01),还可以显著降低血清VLDL、LDL、HDL所携带的胆固醇水平(P<0.05,P<0.01),并且能够降低地鼠肝组织TC(P<0.01,P<0.05)、TG的含量(P<0.01,P<0.05),抑制肝指数(P<0.01,P<0.05),改善中切(P<0.01,P<0.05)和高切(P<0.05,P<0.05)全血黏度,脉抒中剂量还可以改善高脂血症金黄地鼠的血管内皮舒张功能。结论:脉抒具有改善高脂血症金黄地鼠血脂、全血黏度和血管内皮舒张功能的作用。
English Summary:
      To observe the effect of Maishu on blood lipid and vascular endothelial function in golden hamster with hyperlipidemia. Methods:The model of golden hamster with hyperlipidemia was induced by a high fat diet and administrated with MaiShu (low dosage:1 g/kg/d,middle dosage:2 g/kg/d,high dosage:14 g/kg/d). Changes of serum TC,TG,HDL-C and non-HDL-C levels were observed after 2,4,8weeks treatment. Serum VLDL,LDL and HDL were separated by FPLC technique after for 8-week treatment and the level of cholesterol in VLDL,LDL,HDL; testing liver index and TC,TG level in liver were tested. The viscosity of whole blood was measured by the automatic blood viscosity meter. The changes of the endothelium-dependent relaxing reaction of golden hamster mesenteric arteriolar induced by acetylcholine was observed to assess the effect of MS on vascular endothelial function. Results:MS high and middle dose have the function to decrease serum TC level of hamster with hyperlipidemia after 2,4,8 weeks treatment (P<0.01,P<0.01,P<0.05,P<0.01,P<0.01,P<0.01) and the TC level in serum VLDL,LDL,HDL (P<0.05,P<0.01). It also can lower the TC ( P<0.01,P<0.05,TG level in hamster liver,inhibit liver index (P<0.01,P<0.05),improve the whole blood visocity of middle cut (P<0.01,P<0.05) and high cut (P<0.05,P<0.05). Middle dose of MS can also improve the vasodilation function of mesenteric arteriolar in golden hamster model with hyperlipidemia. Conclusion:MS has the function of improving blood lipid,whole blood visocity and vascular endothelial relaxation function in hamster with hyperlipidemia.
