      目的:建立单纯型和肾阳虚复合型2种不同变应性鼻炎模型,探讨小青龙汤对2种变应性鼻炎模型Th1/Th2失衡作用的异同。方法:将105只雄性豚鼠随机分7组,每组15只:空白对照组(A组),单纯模型对照组(B1组),小青龙汤干预单纯模型组(B2组),息斯敏干预单纯模型组(B3组),肾阳虚复合模型对照组(C1组),小青龙汤干预肾阳虚复合模型组(C2组),息斯敏干预肾阳虚复合模型组(C3组)。B1、B2、B3组进行单纯变应性鼻炎造模;C1、C2、C3组在单纯变应性鼻炎的模型上进行肾阳虚复合模型造模。造模成功后,模型组进行用药观察,记录各组局部过敏反应;ELISA法(酶联免疫吸附试验法)检测血清中IFN-γ、IL-4、IL-2和IL-10等细胞因子含量;荧光定量PCR法测脾组织中IFN-γ mRNA、IL-4 mRNA、T-bet mRNA、GATA-3 mRNA等转录因子的相对含量。结果:变应性鼻炎单纯模型和肾阳虚复合模型均出现IFN-γ降低(P<0.01),IL-4升高(P<0.01),IFN-γ/IL-4比值降低(P<0.01),IFN-γ mRNA降低(P<0.01),IL-4 mRNA升高(P<0.01),IFN-γ mRNA/IL-4 mRNA比值降低(P<0.01),T-bet mRNA降低(P<0.01),但可使GATA-3 mRNA升高(P<0.01),因而T-bet mRNA/GATA-3 mRNA比值也相对降低(P<0.01)。小青龙汤可使单纯模型IFN-γ升高(P<0.01),IL-4降低(P<0.01),IFN-γ/IL-4比值升高(P<0.01);升高IFN-γ mRNA(P<0.01),降低IL-4 mRNA(P<0.01),使IFN-γ mRNA/IL-4 mRNA比值升高(P<0.01);使T-bet mRNA升高(P<0.01),但可使GATA-3 mRNA降低(P<0.01),从而使T-bet mRNA/GATA-3 mRNA比值也相对升高(P<0.01)。小青龙汤可降低肾阳虚复合模型中IL-4的含量(P<0.01),对IFN-γ作用不明显,升高IFN-γ/IL-4比值(P<0.01);可使IFN-γ mRNA升高(P<0.01),但对IL-4 mRNA变化不大,IFN-γ mRNA/IL-4 mRNA比值升高(P<0.01);对T-bet mRNA作用不明显,但可使GATA-3 mRNA降低(P<0.05),因而T-bet mRNA/GATA-3 mRNA比值也相对升高(P<0.01)。结论:小青龙通过调节IFN-γ/IL-4比值、IFN-γ mRNA/IL-4 mRNA比值和T-bet mRNA/GATA-3 mRNA比值,来调节Th1/Th2平衡,对单纯变应性鼻炎模型的过敏反应的改善更为明显。
English Summary:
      To establish the simple type of allergic rhinitis model and the complex type of kidney yang deficiency of allergic rhinitis model,and to study the different effects of Xiaoqinglong decoction on the two models of allergic rhinitis,evaluating the difference functions of correcting the imbalance of Th1/Th2.Methods:The simple type of allergic rhinitis model was induced with ovalbumin (OVA) in guinea pigs.Based on this,the complex type of kidney yang deficiency of allergic rhinitis model was established with hydroxyurea administration.Both of the two models were evaluated by observing the weight,survival state and local allergic reaction of guinea pigs,testing the contents of INF-γ,IL-4,IL-2 and IL-10 cytokine in serum by ELISA (enzyme-linked immunosorbent assay),and carrying on real-time fluorescence quantitative PCR method for measuring the relative content of IFN-γ mRNA,IL-4 mRNA,T-bet mRNA and GATA-3 mRNA in spleen tissue.Results:In all models,IFN-γ was reduced,IL-4 increased and the ratio of IFN-γ/IL-4 reduced.In the simple type of allergic rhinitis model,Xiaoqinglong decoction could increase IFN-γ and reduce IL-4,increasing the ratio of IFN-γ/IL-4.As to the complex type of kidney yang deficiency of allergic rhinitis model,the effect on IFN-γ was not obvious.Xiaoqinglong decoction could reduce the IL-4 so that the ratio of IFN-γ/IL-4 of both was reduced,as well as IFN-γ mRNA and IL-4 mRNA was increased.Xiao Qinglong decoction could increase IFN-γ mRNA and decrease IL-4 mRNA in both Simple type of allergic rhinitis model and complex type of kidney yang deficiency of allergic rhinitis model,so that it made the ratio of IFN-γ mRNA/IL-4 mRNA increased,and the role in simple type of allergic rhinitis model was more obvious.Both the simple type of allergic rhinitis model and the complex type of kidney yang deficiency of allergic rhinitis model were found T-bet mRNA decreased,GATA-3 mRNA increased,and the ratio of T-bet mRNA/GATA-3 mRNA decreased.Xiaoqinglong decoction could increase T-bet mRNA and reduce the GATA-3 in the simple type of allergic rhinitis model and thus the ratio of T-bet mRNA/GATA-3 mRNA was increased.In the complex type of kidney yang deficiency of allergic rhinitis model,Xiaoqinglong decoction had no obvious effect on T-bet mRNA,but it also could reduce the GATA-3 mRNA,increasing the ratio of T-bet mRNA/GATA-3 mRNA.Conclusion:Xiaoqinglong decoction can significantly improve the local allergic reaction of the simple type of allergic rhinitis model,but has relatively small effect on the complex type of kidney yang deficiency of allergic rhinitis model.Xiaoqinglong decoction regulates the balance of Th1/Th2 by increasing the ratio of IFN-γ mRNA/IL-4 mRNA and T-bet mRNA/GATA-3 m RNA.Also,it has more obvious effect on the simple type of allergic rhinitis model.
