To observe the hypoglycemic effect of Folium Mori,and analyze its effects on the gene expression of toll-like receptor and its downstream signaling element in the liver from type 2 diabetic rats,and to explore its action mechanismo of hypoglycemic.Methods:The rats were fed with high-fat fodder for 8 weeks,and then were given intraperitoneal injection (ig) with streptozotocin (STZ) to establish the T2DM rats model.The diabetic rats were divided into model group,metformin (0.2 g/kg) group,Folium Mori high and low dose groups (3 g/kg,1 g/kg),with 12 rats in each group,and another 10 rats were selected as the normal control group.The drugs were given by ig administration for 12 weeks,and the fasting blood glucose was detected every week.Oral glucose tolerance test (OGTT) and insulin tolerance test (ITT) were conducted in the mid and later phases of experiment.After the experiment,serum insulin,TNF-,IL-6 and CRP were detected,and the expression level of TLRs and MyD88,TRAF-6,NF-B in the liver were detected by real-time PCR method.Results:After the intervention by Folium Mori,the amount of blood glucose and water intake were decreased significantly (P<0.05 or P<0.01),the glucose tolerance was improved,the contents of IL-6 and CRP in serum were decreased significantly (P<0.05).The expressions of TLR7,TLR8,TLR9 and TRAF-6,MyD88,NF-B were decreased obviously (P<0.05 or P<0.01).Conclusion:Folium Mori can ameliorate diabetes,and its mechanism may be by inhibiting the expression of TLRs and its downstream signal in liver tissue.
