目的:探讨八味茵术颗粒剂中6,7-二甲氧基香豆素、白术内酯Ⅲ、白术内酯Ⅰ的含量测定方法。方法:选取DIKMA Diamonsil C18(2)色谱柱(4.6 mm×250 mm,5 μm),柱温30 ℃,DAD检测(200~400 nm):6,7-二甲氧基香豆素的检测波长为340 nm,白术内酯Ⅲ的检测波长为220 nm,白术内酯Ⅰ的检测波长为275 nm。进样量为5 μL,流动相为水(A)-乙腈(B)梯度洗脱,流速为1 mL/min。结果:6,7-二甲氧基香豆素在0.1~100.0 μg/mL范围内线性关系良好(R2=1.0000)、白术内酯Ⅲ在0.1~100.0 μg/mL范围内线性关系良好(R2=1.0000)、白术内酯Ⅰ在0.1~100.0 μg/mL范围内线性关系良好(R2=1.0000),其精密度、稳定性、加样回收率均符合含量测定要求。结论:本方法简单易行,准确可靠,可有效用于八味茵术颗粒剂的质量控制。 |

To establish the HPLC method for determining the contents of 6,7-dimethoxycoumarin,atractylenolide III and atractylenolide I in Bawei Yinzhu Granules. Methods:The DIKMA Diamonsil C18 (2) (4.6 mm×250 mm,5 μm) column was adopted to separate the sample at the column temperature of 30 ℃. The detection wavelength was set between 200 nm to 400 nm. The detection wavelength of 6,7-dimethoxycoumarin,atractylenolide III and atractylenolide I were 340 nm,220 nm and 275 nm respectively. The injection volume was 5 μL. The mobile phase consisted of water-acetonitrile with a gradient elution at a flow rate of 1.0 mL/min. Results:The calibration curves of were linear in the ranges of 0.1-100.0 μg/mL for 6,7-dimethoxycoumarin (R2=1.0000); the calibration curves were linear in the ranges of 0.1-100.0 μg/mL for atractylenolide III (R2=1.0000); and the calibration curves were linear in the ranges of 0.1-100.0 μg/mL for atractylenolide I (R2=1.0000). The precision,accuracy and recovery met the determination requirements. Conclusion:The method is simple and feasible,and thus can be used for the quality control of Bawei Yinzhu Granules. |
