      目的:观察天麻钩藤汤自发性高血压模型大鼠的治疗作用,并观察其对血管氧化应激的影响。方法:选取SD大鼠100只并随机分为假手术组(10只)、模型组及中药组(共90只),对模型组及中药组大鼠进行高盐饲料喂养以诱导高血压模型,最终将30只造模成功大鼠纳入研究,即模型组、高剂量组及低剂量组,每组10只,中药组大鼠分别接受1.32 kg/mL、及0.33 kg/mL天麻钩藤汤灌胃,1次/d,连续干预10 d,疗程结束后比较4组大鼠血压变化,胸主动脉的病理变化,外周血一氧化氮(NO)、内皮素-1(ET-1)、血管假性血友病因子(vWF)水平变化,并用活性氧(ROS)检测4组胸主动脉细胞氧化应激水平。结果:干预结束后中药组大鼠的血压水平均低于模型组,其中随着天麻钩藤汤剂量升高大鼠收缩压降低越明显,差异有统计学意义(P<0.05);与假手术组比较,模型组及中药组大鼠胸主动脉均出现不同程度的病理受损变化,其中中药组结构、厚度、内膜完整性均较模型组改善,其改善程度高剂量组优于低剂量组。与模型组比较,中药组大鼠外周血ET-1及vWF均有所下降,NO水平有所上调,差异有统计学意义(P<0.05),其中随着天麻钩藤汤剂量增加,大鼠上述指标改善越明显,差异有统计学意义(P<0.05)。与模型组大鼠比较,中药组大鼠胸主动脉ROS明显下降,差异有统计学意义(P<0.05),其中高剂量组下降趋势最明显,与低剂量组比较,差异有统计学意义(P<0.05)。结论:天麻钩藤汤可有效治疗高血压,可抑制动脉氧化应激水平,改善高血压血管重构。
English Summary:
      To observe the therapeutic effects of Tianma Gouteng Decoction on spontaneously hypertensive rats and its effects on the oxidative stress of vascular wall. Methods:A total of 100 SD rats were randomly divided into the sham operation group(10 rats),model group and traditional Chinese medicine(TCM)group(90 rats). The rats in the model group and TCM group were fed with high salt diet to induce hypertensive model. Finally,30 successful SD rats were included in the study,that is,model group,high dose group and low dose group with 10 rats in each group. Rats of TCM group were given Tianma Gouteng Decoction(1.32 Kg/mL)and Tianma Gouteng Decoction(0.33 Kg/mL)once a day for 10 days. The changes of blood pressure,pathological changes of thoracic aorta,levels of nitric oxide(NO),endothelin-1(ET-1)and von Willebrand factor(vWF)in peripheral blood of rats in each group were compared after treatment. The level of cell oxidative stress in thoracic aorta wall of each group was detected. Results:1)After the intervention,the blood pressure level of rats in the TCM group was lower than that of the model group. The systolic blood pressure of rats in the Tianma Gouteng Decoction group decreased significantly with the increase of the dosage. The difference was statistically significant(P<0.05); 2)HE staining showed that:compared with the sham operation group the thoracic aorta of rats in the model group and the TCM group appeared different impaired physiology. The structure,thickness and intimal integrity of the TCM group were better than those of the model group. The degree of improvement in the high dose group was higher than that of the low dose group. 3)Compared with the model group,ET-1 and vWF in peripheral blood of rats in TCM group decreased,while NO concentration was up-regulated. The difference was statistically significant(P<0.05). With the increase of Tianma Gouteng Decoction dosage,the previous indexes of rats were improved more significantly. The difference was statistically significant(P<0.05). 4)Compared with the model group,the ROS of thoracic aorta of rats in the TCM group decreased significantly. The difference was statistically significant(P<0.05). The trend of ROS in the high-dose group was the most obvious(P<0.05). The difference was statistically significant. Conclusion:Tianma Gouteng Decoction can effectively treat hypertension,inhibit arterial oxidative stress as well as improve vascular remodeling of hypertension.
