      目的:优化构建抗抑郁研究的细胞实验模型,探讨西红花对皮质酮(CORT)诱导的PC12细胞损伤的保护作用及其相关机制。方法:观察不同浓度(0、125、250、500、750、1 000 μmol/L)皮质酮损伤PC12细胞24 h、36 h、48 h,CCK-8检测细胞活力,优化构建抑郁体外模型;不同浓度(2.5、5、10 μg/mL)西红花预处理PC12细胞24 h,CCK-8检测细胞活力,观察西红花对CORT(500 μmol/L)细胞损伤后的保护作用;检测细胞培养上清中的LDH活性,判断药物对细胞膜的保护作用;使用Annexin V-FITC/PI双染色法检测药物对CORT诱导的PC12细胞凋亡的保护作用;检测细胞裂解液中Caspase-3活性,判断药物作用与Caspase蛋白通路相关性。结果:CORT(0~1 000 μmol/L)作用PC12细胞,细胞存活率随药物浓度的增加而逐渐降低,依剂量呈线性关系;当西红花浓度>100 μg/mL时,对PC12细胞存活的影响出现统计学意义;西红花预处理后,细胞存活率由模型组(53.31±4.18)%,上升为(57.46±2.76)%、(60.48±2.73)%、(61.99±3.05)%、(61.38±2.26)%、(59.46±2.41)%、(58.64±3.74)%;西红花预处理组LDH释放量显著低于模型组(P<0.05);西红花能够明显抑制CORT诱导的PC12细胞凋亡,西红花可抑制Caspase-3的表达。结论:西红花对CORT诱导的PC12细胞有一定保护作用。
English Summary:
      To optimized the establishment of a model of depression in vitro,and to explore the protective effects of Crocus sativus L on PC12 cell injury induced by CORT and its related mechanisms.Methods:PC 12 cells were treated with CORT in different concentrations (0,125,250,500,750 and 1 000 μmol/mL) for 24,36 and 48 h to establish a depression model in vitro.PC12 cells were pretreated with Crocus sativus L at different concentrations (2.5,5 and 10 ug/mL) for 24 hours.Cell viability was measured by CCK-8,and the protective effects of Crocus sativus L on PC12 cell injury induced by CORT were determined.The levels of LDH activity in supernatant were detected to judge the protection effect of the drugs on PC12 cells.The protective effect on PC12 apoptosis was analyzed by Annexin V-FITC/PI staining.Caspase-3 activity was detected in cell lysates and correlation between drug action and Caspase protein pathway was judged.Results:After CORT (0-1 000 μmol/L) treatment of PC12 cells for 24 h,the cell survival rates of each concentration were decreased with dose increasing in a dose-dependent manner.When the concentration of Crocus sativus Lextract was greater than 100 μg/mL,the effect on PC12 cells was statistically significant.After drug pretreatment,the PC12 cells survival rate was increased from model group (53.31±4.18)% to (57.46±2.76)%,(60.48±2.73)%,(61.99±3.05)%,(61.38±2.26)%,(59.46±2.41)% and (58.64±3.74)%,respectively.The release of LDH from Crocus sativus L pretreated group was significantly lower than that of model group (P<0.05).Crocus sativus L can significantly inhibit CORT-induced apoptosis of PC12 cells.Crocus sativus Lcan inhibit the expression of Caspase-3.Conclusion:Crocus sativus L has the protective effects on PC12 cell injury induced by CORT.
