Abstract:To observe the antipyretic effect of Wuhua tea fever model rats,to further clarify its antipyretic mechanism, and to study its effect on liver and kidney function. Methods:A total of 50 SPF SD rats were randomly divided into blank group, model group and tea group. The febrile model was made by dry yeast in the model group and the Wuhua tea group, and then the rats in the Wuhua tea group were given high, middle and low doses [6480 g/(kg·d),3240 g/(mol·d)and 1620 g/(mol·d)],respectively.The blank group and the model group received the same volume of normal saline for gavage. Each group was treated with continuous intervention for 5 days. The body temperature, liver and kidney function, peripheral serum interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),IL-1 β,TNF-α,TNF-β,TNF-α,TNF-β in peripheral serum were observed. The expression level of cyclooxygenase 1(COX-1)changed.Results:1)After modeling, the body temperature of the rats was significantly higher than that of the blank group(P<005),and the body temperature showed bimodal heat type, which appeared at 2 h and 5 h after operation, respectively. In the course of the study, some of the rats showed obvious mental malaise, dark hair color, and even shivering symptoms, which indicated that the model was successful.Compared with the model group, the body temperature of rats in the Wuhua tea group decreased significantly(P<005),and there was no significant difference in antipyretic effect among the intervention groups(P>005)2)Compared with the blank group, there was no significant difference in ALT,AST,BUN and Cr between the model group and the five-flower tea group(P>005).In the five-flower tea dry preparation group, the intestinal propulsive rate in the five-flower tea+astragalus membranaceus group and the five-flower tea+jujube group was lower than that in the five-flower tea group. The difference was statistically significant(P<005).The intestinal propulsive rate of Wuhua tea+jujube+astragalus membranaceus group was lower than that of the other two groups, the difference was statistically significant(P<005)3)Compared with the blank group,IL-6,TNF-α and IL-1 β in the model group and the tea group were significantly increased(P<005),and the changes of the above indexes in the tea group were more obvious than those in the model group. The change range of Wuhua tea+jujube+astragalus membranaceus group was the largest(P<005)4)Western blotting test showed that compared with the blank group, the COX-1 in the hypothalamus of the model group and the tea group decreased significantly(P<005),and the concentration of COX-1 in the group of the five flowers tea increased significantly(P<005),and the concentration of MDA in the hypothalamus of the model group and the tea group increased significantly(P<005).There was no significant difference in OD value of COX-1 among the three groups(P>005).Conclusion:Wuhua tea has obvious antipyretic effect in a dose-dependent manner and has no hepatorenal toxicity.it can be popularized and applied in clinic.
