      目的:探讨养血清脑颗粒对认知功能障碍大鼠学习、记忆及GAP-43、PSD-95表达的影响。方法:采用双侧颈动脉永久性结扎法构建认知功能障碍大鼠模型,将大鼠随机分为模型组与实验组,每组18只。假手术组(n=15)大鼠仅分离、暴露颈动脉,不结扎。实验组灌胃养血清脑颗粒10 mL/(kg·d),假手术组与模型组灌胃等量生理盐水,连续干预4周。测定各组大鼠的学习记忆能力;HE染色观察大鼠海马组织病理学变化;TUNEL检测海马神经元凋亡情况;Wb法检测海马组织GAP-43和PSD-95蛋白表达。结果:与假手术组比较,模型组大鼠逃避潜伏期与游泳路程延长,跨越原平台次数减少,记忆成绩下降,神经元凋亡指数与海马组织PSD-95蛋白相对表达量升高,而GAP-43蛋白相对表达量降低,且以上指标实验组均优于模型组(均P<0.05)。结论:养血清脑颗粒可改善认知功能障碍大鼠的学习记忆能力,抑制神经元凋亡,其机制可能与上调海马组织GAP-43蛋白表达,下调PSD-95蛋白表达相关。
English Summary:
      To investigate the effects of Yangxue Qingnao Granules on learning,memory and expression of GAP-43 and PSD-95 in rats with cognitive impairment.Methods:A bilateral carotid artery permanent ligation was used to construct the rat models of cognitive dysfunction,and the rats with cognitive impairment were randomly divided into a model group and a test group,with 18 rats in each group.Rats in the sham-operation group(n=15)only isolated and exposed the carotid arteries without ligation.Rats in the test group were administered with Yangxue Qingnao Granules 10 mL/(kg·d) by gavage.The sham-operation group and the model group were perfused with the same amount of normal saline,and the intervention was continued for 4 weeks.The learning and memory ability of rats were detected in each group; histopathological changes in hippocampus of rats by HE staining was observed; TUNEL was used to detect hippocampal neuron apoptosis in each group of rats; Wb method was used to detect the protein expression of GAP-43 and PSD-95 in hippocampus of rats.Results:Compared with the sham-operation group,the escape latency and swimming distance of the model group were significantly longer,the number of times to cross the original platform significantly reduced,the memory score significantly decreased,the apoptosis index of hippocampal neurons and the relative expression of PSD-95 protein in the hippocampus significantly increased in the model group,but the relative expression of GAP-43 protein significantly reduced,and the above indicators were better in the test group than in the model group(all P<0.05).Conclusion:Yangxue Qingnao Granules can improve the learning and memory ability,inhibit neuronal apoptosis,and its mechanism may be related to up-regulating the expression of GAP-43 protein and down-regulating the expression of PSD-95 protein in hippocampus.
