To investigate the effects and related mechanism of blueberry anthocyanins (BA) on the proliferation and invasion of human gastric adenocarcinoma BGC-823 cells.Methods:Human gastric adenocarcinoma BGC-823 cells exp-L/M/H and the control group were treated with BA at the concentration of 25,50,100 μg/mL and the same amount of normal saline respectively,and each group was treated for 12,24,and 48 hours.The proliferation and apoptosis of human gastric adenocarcinoma BGC-823 cells was detected by MTT assay and Flow cytometry; Transwell chamber assay and immunocytochemistry method were used to detect the invasion of human gastric adenocarcinoma BGC-823 cells and the expression of P53 and caspase-3 protein.Results:At 48 h after intervention,the percentage of P53 cells and Caspase-3 protein positive cells in human gastric adenocarcinoma BGC-823 cells in the exp-L/M/H were (11.12±1.01)%,(24.31±2.18)%,(47.56±2.91)%,(66.49±5.14)% and (15.04±1.23)%,(28.22±3.09)%,(69.36±5.42)%,and (86.52±6.15)%,respectively.With the increase of BA concentration,the proliferation inhibition rate of gastric adenocarcinoma BGC-823 cells in experimental group increased gradually; Compared with the control group,the apoptotic rate,P53 protein and Caspase-3 protein positive percentage of human gastric adenocarcinoma BGC-823 cells in the exp-L/M/H groups significantly increased,while the invasive rate decreased and the trend was concentration-dependent (all P<0.05).Conclusion:BA can effectively inhibit the proliferation of gastric adenocarcinoma BGC-823 cells,induce its apoptosis,and reduce its invasive ability,which may be related to the up-regulation of P53 protein and Caspase-3 protein expression.
