    目的:观察参蒲盆炎颗粒对苯酚胶浆致子宫内膜炎大鼠模型的影响,探讨其抗炎作用机制。方法:选取SD大鼠60只并随机分为假手术组、模型组、参蒲盆炎颗粒高、中、低剂量组(15.0,7.5,3.75 g生药/kg)和妇科千金胶囊组(5.0 g生药/kg)。采用苯酚胶浆复制子宫内膜炎大鼠模型,连续灌胃给药21 d,比色法测定大鼠血清中超氧化物歧化酶(SOD)、丙二醛(MDA)水平,HE染色评价大鼠子宫内膜的病理变化,酶联免疫吸附试验法(ELISA)检测子宫组织肿瘤坏死因子(TNF-α)及白细胞介素-1β(IL-1β)的水平,蛋白免疫印迹法(WB)检测子宫组织中核因子-κB(NF-κB)的表达。结果:参蒲盆炎颗粒能明显改善子宫内膜上皮细胞增生、变性、坏死及炎细胞浸润等病理改变;参蒲盆炎颗粒高、中剂量组显著升高大鼠血清SOD水平及降低MDA水平;参蒲盆炎颗粒显著降低大鼠子宫组织IL-1β、TNF-α水平及核因子-κB蛋白的磷酸化水平。结论:参蒲盆炎颗粒对苯酚胶浆致子宫内膜炎大鼠有较好的治疗作用,其作用机制可能与其抑制核因子-κB通路激活有关。

    Abstract:

    To observe the effects of Shenpu Penyan Granules on endometritis induced by phenol mucilage and the explore anti-inflammatory mechanism.Methods:A total of 60 female SD rats were randomly divided into a sham operation group,a model group,a Shenpu Penyan Granules high dose group,a medium dose group and a low dose group (15.0,7.5,3.75 crude drugs g/kg) and Fuke Qianjing Capsule (5.0 g/kg).Rat model of endometritis was copied with phenol glue.After the rats were treated 21 days by intragastric administration,the content of superoxide dismutase (SOD) and malondialdehyde (MDA) in serum of rats were determined by colorimetry.Pathological changes of endometrium of rates were evaluated by rats.Expressions of NF-kB in uterine tissue were detected by western blotting (WB).Results:Shenpu Penyan Granules could improve obviously the pathological damage of endometrial epithelial cell proliferation,degeneration,necrosis and inflammatory cell infiltration.Shenpu Penyan Granules high and medium dose group significantly increased the serum SOD content and decreased MDA content.And Shenpu Penyan Granules significantly reduced the content of IL-1β,TNF-a and NF-kB protein phosphorus level in rat uterine tissue.Conclusion:Shenpu Penyan Granules have a good therapeutic effects on phenol-induced endometritis in rats and its mechanism may be related to the inhibition of NF-kB pathway activation.
