      目的:探讨补肾活血汤对股骨头坏死的疗效,同时观察其对Wnt-βcatenin信号通路的影响。方法:将45只SD大鼠随机分为空白组、模型组及中药组,每组15只。利用甲泼尼龙琥珀酸钠制备激素性股骨头坏死动物模型,成模后中药组大鼠接受补肾活血汤灌胃,1次/d,空白组及模型组大鼠接受生理盐水灌胃,均连续灌胃6周,比较各组大鼠股骨形态学、骨密度、股骨头病理学、血清钙、磷、股骨头组织Wnt10b、β-Catenin水平变化。结果:1)模型组大鼠股骨头密度高低不同,股骨头毛糙,骨小梁模糊,中药组大鼠股骨头骨小梁数目较模型组增多,且相对清晰,股骨头面较模型组光滑。2)模型组及中药组大鼠骨密度、血清钙、磷水平、股骨头组织Wnt10b、β-Catenin蛋白均明显下降(P<0.05),中药组高于模型组(P<0.05)。3)模型组大鼠股骨头软骨出现明显病理改变;中药组大鼠股骨头结构尚完整,空骨陷窝较模型组减少,软骨细胞排列欠规则。结论:补肾活血汤对股骨头坏死有保护效应,其作用机制与Wnt-βcatenin信号通路有关。
English Summary:
      To explore the therapeutic effect of Bushen Huoxue decoction on osteonecrosis of the femoral head and its effect on Wnt-β catenin signal pathway. Methods:A total of 45 SD rats were randomly divided into blank group (n=15), model group (n=15) and traditional Chinese medicine group (n=15). The animal model of steroid-induced osteonecrosis of the femoral head was made by methylprednisolone sodium succinate. the rats in the traditional Chinese medicine group were fed with Bushen Huoxue decoction once a day, and the rats in the blank group and model group were fed with normal saline for 6 weeks. The changes of femoral morphology, bone mineral density, pathology of femoral head, serum calcium, phosphorus, Wnt10b and β-Catenin in each group were compared. Results:1) the density of the femoral head of the model group was different, the femoral head was rough and the trabeculae were blurred. The number of trabeculae of the femoral head in the traditional Chinese medicine group was more than that in the model group, and it was relatively clear, and the femoral head in the traditional Chinese medicine group was smoother than that in the model group. 2) the bone mineral density (BMD), serum calcium and phosphorus levels, Wnt10b and β-Catenin protein in femoral head tissue of rats in model group and traditional Chinese medicine group were all decreased significantly (P<0. 05). It was higher in the traditional Chinese medicine group than that in the model group (P<0. 05). 3) there were obvious pathological changes in the cartilage of the femoral head in the model group, the structure of the femoral head in the traditional Chinese medicine group was still intact, the empty lacunae was less than that in the model group, and the arrangement of chondrocytes was irregular. Conclusion:Bushen Huoxue decoction has protective effect on osteonecrosis of femoral head, and its mechanism is related to Wnt-β catenin signal pathway.
