      目的:应用网络药理学研究方法预测复方紫草油治疗肛周湿疹的有效成分,作用靶点及其潜在的作用机制。方法:通过检索中药系统药理学平台(TCMSP)和中药综合数据库(TCMID)筛选出复方紫草油的有效成分和作用靶点;通过检索人类基因信息数据库(GeneCards)得到肛周湿疹的疾病靶点;通过R语言走脚本得到药物和疾病共同作用靶点;经过Cytoscape软件构建药物-有效成分-作用靶点-疾病的互作网络,探索关键有效成分及靶点;运用STRING平台构建蛋白质间相互作用网络(PPI)进行可视化分析;使用Bioconductor生物信息软件包进行GO功能富集分析和KEGG通路分析。结果:筛选后共得出45个药物有效成分和相应靶点409个,并得到189个疾病相关作用靶点,其中有14个是药物与疾病的共同靶点。富集分析共得到31个GO功能条目和73条KEGG信号通路。结论:复方紫草油治疗肛周湿疹可能是通过IL-6、MMP9、CXCL8、ICAM1和STAT3等蛋白作用,发生细胞因子受体结合、转录因子及核受体活性等生物学过程,并主要富集在African trypanosomiasis、Hepatitis B、AGE-RAGE等多条信号通路上,发挥抗炎和修复屏障等作用。
English Summary:
      To predict the effective components,action targets and potential mechanisms of compound lithospermum oil in the treatment of perianal eczema by network pharmacology.Methods:The effective ingredients and targets of compound oleum lithospermi were screened out by searching Pharmacology Platform of Traditional Chinese Medicine System(TCMSP)and Chinese Medicine Comprehensive Database(TCMID).Human gene information database(GeneCards)was used to select the disease therapeutic targets of perianal eczema.R language was used to run the script to obtain the target of the drug and the disease; Cytoscape software was used to construct the drug-active ingredient-action target-disease interaction network to explore the key active ingredients and targets; the STRING platform was used to build the protein interaction network(PPI)for visual analysis; Bioconductor bioinformatics software package was used for GO function enrichment analysis and KEGG pathway analysis.Results:After screening,a total of 45 drug active ingredients and 409 corresponding targets were obtained.189 disease-related targets were obtained,of which 14 were common targets for drugs and diseases.After enrichment analysis,31 GO functional entries and 73 KEGG signal pathways were obtained.Conclusion:Compound oleum lithospermi in the treatment of perianal eczema may be through the action of IL-6,MMP9,CXCL8,ICAM1,STAT3 and other proteins,resulting in the biological process of cytokine receptor binding,cytokine activity and nuclear receptor activity.These targets mainly concentrated in multiple signaling pathways such as African trypanosomiasis、Hepatitis B、AGE-RAGE,which play key roles of anti-inflammatory and repair barrier.
