      目的:探讨黄连素对多囊卵巢综合征(PCOS)大鼠卵巢血供的影响。方法:30只大鼠随机分为模型组、实验组和阳性对照组,采用脱氢表雄酮(DHEA)皮下注射构建PCOS大鼠;另选10只仅皮下注射大豆油作为对照组,1次/d,连续注射20 d。实验组和阳性对照组分别灌胃黄连素(80 mg/kg)和罗格列酮(3 mg/kg),模型组和对照组灌胃生理盐水(10 mL/kg),1次/d,连续干预22 d。检测大鼠血清睾酮(T)和游离睾酮(FT)含量,苏木精-伊红(HE)染色观察大鼠卵巢组织形态及微血管密度(MVD),原位杂交法与免疫组化法分别检测大鼠卵巢组织血管内皮生长因子(VEGF)mRNA水平与甾体类生成快速调节蛋白(StAR)蛋白表达。结果:模型组大鼠卵巢组织MVD、VEGF mRNA表达及StAR蛋白相对表达量较对照组升高,实验组和阳性对照组较模型组降低,且实验组低于阳性对照组(P<0.05)。结论:黄连素可通过调控PCOS大鼠卵巢组织血供及血管生成而改善卵巢组织形态,同时可通过下调StAR蛋白表达改善高雄激素血症。
English Summary:
      To investigate the effects of berberine on ovarian blood supply in rats with polycystic ovarian syndrome (PCOS).Methods:A total of 30 rats were randomly divided into a model group,a test group and a positive control group.Dehydroepiandrosterone (DHEA) subcutaneous injection was used to construct PCOS rats; another 10 rats were selected as the control group by subcutaneous injection of soybean oil,all were intervented once a day for 20 d continuously.The test group and positive control group were given by gavage with berberine 80 mg/kg and rosiglitazone 3 mg/kg respectively.The model group and control group were given 10 mL/kg normal saline by gavage,all were given once a day for consecutive 22 d.The content of testosterone (T) and free testosterone (FT) in serum of rats were detected.Hematoxylin-eosin (HE) staining was used to observe rat ovarian tissue morphology and microvessel density (MVD).In situ hybridization and immunohistochemistry were used to detect vascular endothelial growth factor (VEGF) mRNA levels,producing rapid regulatory protein (StAR) protein expression with steroids.Results:The expression of MVD and VEGF mRNA and the relative expression of StAR protein in the ovarian tissue of the model group were higher than those of the control group,and the test group and the positive control group were lower than the model group.The experimental group was lower than the positive control group (P<0.05).Conclusion:Berberine can improve ovarian morphology by regulating blood supply and angiogenesis in ovarian tissues of PCOS rats; and can improves hyperandrogenism by down-regulating the expression of StAR protein.
