      目的:根据网络药理学方法对五苓散治疗糖尿病肾病(DKD)的作用机制进行研究探讨。方法:利用中药系统药理学数据库与分析平台(TCMSP)对五苓散中有效成分进行筛选及靶点预测,检索GeneCards、CTD数据库筛选DKD疾病靶点,利用韦恩图在线平台筛选五苓散治疗DKD交集靶点,借助网络拓扑分析插件CytoNCA筛选五苓散治疗DKD核心靶点,使用Cytoscape 3.7.1建立化合物-疾病-靶点调控网络,基于DAVID数据库对核心靶点进行GO功能富集和KEGG通路富集分析。结果:基于TCMSP得到五苓散的27个有效成分和62个潜在靶点,筛选获得与DKD疾病发生、发展相关的靶点21 153个,借助网络拓扑分析插件CytoNCA筛选五苓散治疗DKD核心靶点16个,GO功能富集分析得到条目99个,其中生物过程条目64个、分子功能条目24个、细胞组成条目11个,主要包括过脂质反应、G蛋白偶联受体信号途径、肾上腺素能受体活性、四吡咯结合、胞膜小凹等功能途径;KEGG通路富集分析得出条目61个,主要包括NF-κB信号通路、细胞凋亡、TNF信号通路、雌激素信号途径、IL-17信号通路等通路途径。结论:基于网络药理学初步探讨并验证了五苓散治疗DKD多成分、多靶点、多通路的整体调节作用特点,预测了五苓散治疗DKD的潜在作用机制,以期为其活性成分研究与实验研究提供科学依据。
English Summary:
      To explore the mechanism of Wuling Powder(WLP)in treatment of diabetic kidney disease(DKD)based on network pharmacology.Methods:Using TCMSP to screen effective components and targets of WLP,searching GeneCards,CTD to screen targets of DKD,constructing Venn online platform to get intersection targets of WLP-DKD,selecting common targets of WLP-DKD by using CytoNCA,using Cytoscape 3.7.1 to build component-disease-target network,analyzing GO function enrichment and KEGG pathway enrichmen by David database.Results:27 effective components and 62 potential targets of WLP were obtained based on TCMSP,21 153 targets related to the occurrence and development of DKD were obtained,16 common targets of WLP-DKD were obtained by using CytoNCA.99 items were obtained by GO function enrichment analysis,including 64 biological process items,24 molecular function items and 11 cellular component items,mainly including response to lipid,G protein-coupled receptor signaling pathway,adrenergic receptor activity,tetrapyrrole binding,caveola.KEGG pathway enrichment analysis obtained 61 items,including TNF signaling pathway,IL-17 signaling pathway,NF-κB signaling pathway,apoptosis and estrogen signaling pathway signaling pathway.Conclusion:Based on network pharmacology,the overall regulatory characteristics of WLP in treatment of DKD were discussed and verified,and the potential mechanism of WLP in treatment of DKD was predicted,in order to provide scientific basis for the study of active ingredients and experimental research.
