      目的:体质在疾病的形成、发展和转归中起着重要作用。本研究从转录组学水平首次初步揭示阴虚体质者外周血中lncRNA和mRNA的表达谱及相关作用网络。方法:运用Agilent Human lncRNA+mRNA(4×180 k)表达谱芯片,从阴虚体质(n=10)和平和体质(n=9)者的外周血中分离单个核细胞(PBMCs),采用“limma”方法筛选2组的差异基因,然后对其进行GO功能和IPA通路的显著性富集分析,并寻找lncRNA近距离调控的靶基因。结果:差异表达的lncRNAs和mRNAs均能够将阴虚体质和平和体质进行区分。阴虚体质中有显著差异表达的lncRNAs 829个(282个上调,547个下调)和mRNAs 1 435个(1 038个上调,397个下调)。GO功能富集和IPA作用通路结果提示,这些差异表达基因主要与免疫调节、代谢紊乱和信号传导失常通路显著相关。例如,T细胞受体信号转导,NF-κB信号通路,L-arabinose代谢过程和I型糖尿病信号传导等。确定了7个lncRNA-mRNA相互作用对,比如,LTA-HCP5、ORAI3-AC135048.13和BCL7C-AC135048.13等。结论:阴虚体质易感干眼症、高血压、糖尿病等疾病的物质基础可能与lncRNAs/mRNAs的免疫调节、代谢作用和信号传导失常相关。
English Summary:
      Constitution in Traditional Chinese Medicine(TCM)plays an important role in the initiation,development,and prognosis of diseases.We aim to identify a previously unexplored lncRNA and mRNA expression signatures and lncRNA-mRNA regulatory networks in the Yin Deficiency Constitution(YDC)from the transcriptome.Methods:lncRNA+mRNA Human Gene Expression Microarray V4.0(4×180 K)was performed in this study.The Peripheral Blood Mononuclear Cells(PBMCs)were isolated from the subjects with YDC(n=10)and Balanced Constitution(BC)(n=9).The “limma” method was performed between groups of significantly differentially expressed lncRNAs and mRNAs,The regulated lncRNA target genes were investigated by bioinformatics techinques, including Gene Ontology (GO) and Ingenuity Pathway Analysis(IPA).Results:The differential expression lncRNAs/mRNAs could be used to distinguish individuals with YDC from those with BC.These were 829 DElncRNAs(282 up-regulated,547 down-regulated)and 1435 mRNAs(1 038 up-regulated and 397 down-regulated)in the YDC.GO enrichment analysis and IPA functional indicated that these differentially expressed genes were associated with Immune regulation,metabolic disorders and signal transduction disorders.For example,T cell receptor signaling,NF-κB signaling pathway,L-Arabinose metabolic processes,and type 1 diabetes signaling and so on.Seven most significant interaction pairs of lncRNA-nearby targeted mRNAs were identified including LTA-HCP5,ORAI3-AC135048.13 and BCL7C-AC135048.13.Conclusion:This study revealed that lncRNAs/mRNAs could be used as potential biomarkers in the YDC,and the material basis of YDC susceptibility to xerophthalmia,hypertension,diabetes mellitus might contribute to immunity and hormone regulation.
