      目的:探索榆栀止血颗粒对子宫内膜异位症大鼠磷脂酰肌醇3-激酶/蛋白激酶B(PI3K/Akt)信号通路及异位内膜体积的影响。方法:构建子宫内膜异位症大鼠模型,随机分为模型组、榆栀止血颗粒低、中、高剂量组、孕三烯酮组,每组12只,另取12只设为假手术组,分组处理后处死大鼠,测定大鼠异位子宫内膜体积,以免疫组织化学染色检测大鼠异位子宫内膜组织中微血管密度(CD31阳性细胞表达),以免疫酶联吸附试验(ELISA)检测大鼠血清VEGF、MMP-9、IL-6、TNF-α水平;以蛋白免疫印迹法检测大鼠异位子宫内膜组织PI3K/Akt通路相关蛋白表达。结果:与假手术组比较,模型组大鼠CD31阳性细胞比例、血清VEGF、MMP-9、IL-6及TNF-α水平、异位子宫内膜组织p-PI3K/PI3K、p-Akt/Akt均升高(P<0.05);与模型组比较,榆栀止血颗粒低、中、高剂量组及孕三烯酮大鼠异位子宫内膜体积、CD31阳性细胞比例、血清中VEGF、MMP-9、IL-6及TNF-α水平、异位子宫内膜组织p-PI3K/PI3K、p-Akt/Akt均降低(P<0.05),且榆栀止血颗粒各组之间呈剂量依赖性(P<0.05);榆栀止血颗粒高剂量组与孕三烯酮组比较,差异无统计学意义(P>0.05)。结论:榆栀止血颗粒可抑制子宫内膜异位症模型大鼠内膜血管生成,减轻炎性反应,进而减小其异位内膜体积,可能通过阻滞PI3K/Akt信号传导实现。
English Summary:
      To explore the effects of Yuzhi Zhixue Granules on the signal pathway of phosphatidylinositol 3-kinase/protein kinase B (PI3K/Akt) and the volume of ectopic endometrium in rats with endometriosis.Methods:The rat model of endometriosis was established and randomly divided into a model group,a low,middle,high dose Yuzhi Zhixue Granules group and a gestrinone group,with 12 rats in each group.Another 12 were set as sham operation group,the rats were killed after treatment,the volume of ectopic endometrium of rats was measured,the microvessel density (CD31 positive cell expression) was detected by immunohistochemistry,the levels of VEGF,MMP-9,IL-6 and TNF-α in serum were detected by enzyme linked immunosorbent assay (ELISA); and the expression of PI3K/Akt pathway related proteins was detected by Western blot.Results:Compared with the sham operation group,the proportion of CD31 positive cell,levels of VEGF,MMP-9,IL-6 and TNF-α in serum,the p-PI3K/PI3K and p-Akt/Akt in the ectopic endometrium tissues of the model group were all higher (P<0.05); Compared with the model group,the low,medium and high doses of Yuzhi Zhixue Granules and the ectopic endometrial volume of gestrinone rats,the proportion of CD31 positive cells,serum VEGF,MMP-9,IL-6 and TNF-α level,p-PI3K/PI3K and p-Akt/Akt of ectopic endometrial tissue were all decreased (P<0.05),and there was a dose-dependent relationship between Yuzhi Zhixue Granule groups (P<0.05); there was no significant difference between high dose Yuzhi Zhixue Granule group and gestrinone group (P>0.05).Conclusion:Yuzhi Zhixue Granule can inhibit the angiogenesis of endometrium,reduce inflammation and reduce the volume of endometrium,which may be achieved by blocking PI3K/Akt signal transduction.
