基于网络药理学探析核心中药治疗颈椎病的作用机制

      目的:预测中药抗颈椎病主要活性成分及其作用靶点,探讨其成分-靶点的作用机制。方法:检索近10年内服治疗颈椎病的有效中药复方,应用Access 2003数据库录入复方,应用Apriori算法进行关联分析,运用软件Cytoscape建立中药配伍网络,选择网络相对中心度较高的前5味药作为核心中药,通过中药系统药理学数据库与分析平台(Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform,TCMSP)数据库查询中药的化学成分,运用Uniprot数据库查询靶蛋白对应的Gene Name,应用Cytoscape3.6进行成分-靶标-通路的网络分析和GO分析,通过KOBAS网络富集分析。结果:共纳入复方332首,当归、葛根、黄芪、川芎和甘草构成核心中药;共得到419个疾病靶标以及包括丝裂原活化蛋白激酶(Mitogen-activated Protein Kinase,MAPK)1、MAPK14、CCND1、EGFR、FOS、SRC等10个核心靶标在内的312个药物靶标。根据靶标对应的化学成分,从五味核心中药中筛选出包括槲皮素、大豆苷元、山柰酚、葛根素等146个成分。结论:五味核心中药很可能主要作用于MAPK14、FOS、CXCL8、CCL2、MAPK1、SRC等核心靶标及IL-17通路、MAPK通路,P13k-AKT信号通路、TNF信号通路等关键通路,通过调控细胞凋亡,延缓椎间盘老化,降低炎症介质的表达,减轻疼痛,通过减少缺血性伤害治疗眩晕以及促进钙离子释放,减缓肌肉痉挛来达到治疗颈椎病的目的。
English Summary:
      To predict and discover the main active ingredients and their targets of traditional Chinese medicine(TCM)against cervical spondylosis,and to explore the mechanism of their ingredients and targets.Methods:Effective Chinese medicine compound prescriptions for the treatment of cervical spondylosis in the past 10 years were retrieved and entered into Access 2003 database.Apriori algorithm was used for correlation analysis while the software Cytoscape was used to establish a Chinese medicine compatibility network,where the top 5 medicines with relatively high centrality were selected as the “core herbs”.The chemical compositions of these herbs were screened through the TCMSP database; the Gene Name corresponding to the target protein were searched using the Uniprot database.And then Cytoscape3.6 was used to perform network analysis and GO analysis of the component-target-pathway,which was enriched through the KOBAS website analysis.Results:A total of 332 compounds were included,with Radix Angelicae Sinensis,Radix Puerariae,Radix Astragali seu Hedysari,Rhizoma Ligustici Chuanxiong and Radix Glycyrrhizae as the core herbs of TCM; a total of 419 disease targets and 312 drugs including 10 core targets such as MAPK1,MAPK14,CCND1,EGFR,FOS and SRC target were obtained.According to the chemical composition corresponding to the target,a total of 146 components including quercetin,daidzein,kaempferol and puerarin were screened from the five core herbs.Conclusion:The 5 core herbs are likely to mainly act on MAPK14,FOS,CXCL8,CCL2,MAPK1,SRC and other core targets and key pathways such as IL-17 pathway,MAPK pathway,P13k-AKT signaling pathway,and TNF signaling pathway.By regulating cell apoptosis,it can delay intervertebral disc aging,reduce the expression of inflammatory factors,and reduce pain,reduce the ischemic injury to treat vertigo and promote calcium ion release and slow muscle spasm to achieve the purpose of treating cervical spondylosis.
