目的:运用网络药理学方法探讨当归六黄汤治疗心律失常的物质基础和分子机制。方法:运用网络数据库对当归六黄汤成分靶点和心律失常靶点进行检索筛选;利用Cytoscape3.7.2软件构建中药成分-靶点网络;利用STRING数据库构建当归六黄汤治疗心律失常的蛋白质-蛋白质相互作用(PPI)网络图,经拓扑分析找出核心靶点,最后对药物-疾病交集靶点进行生物通路及富集分析。结果:共收集到当归六黄汤135个有效成分及其2 714个成分靶点;心律失常疾病靶点400个;药物成分和疾病的共有靶点153个;以大于平均Degree值2倍(14.78)进行拓扑分析,最终得到关键核心靶点4个:NR3C1、NOS3、ESR1、PTGS2;GO生物过程和KEGG富集分析显示:该方治疗心律失常主要涉及375个生物过程,34条KEGG途径,生物过程主要涉及细胞质膜、质膜部分、离子束缚、细胞内信号级联、对有机物的反应、细胞增殖的调控等生物过程;其KEGG信号通路主要通过神经活性配体-受体相互作用的可能性最大,其次是钙信号通路、心肌细胞的肾上腺素信号通路传导。结论:当归六黄汤治疗心律失常具有多成分、多靶点、多通路的特点,其机制较为复杂,可能主要通过对神经活性配体-受体相互作用来实现治疗作用。 |

To explore the material basis and molecular mechanism of Danggui Liuhuang Decoction in the treatment of patients with arrhythmia by using network pharmacology.Methods:The online database was used to search and screen the targets of Danggui Liuhuang Decoction and Arrhythmia; Cytoscape 7.2 Software was used to construct the traditional Chinese medicine component-target network; STRING Database was used to construct the protein-protein interaction network diagram(PPI) of Danggui Liuhuang Decoction in the treatment of arrhythmia,and find the core targets through topological analysis.Finally,biological pathways and enrichment analysis were performed on drug-disease intersection targets.Results:A total of 135 active ingredients,2714 component targets of Danggui Liuhuang Decoction; 400 targets of arrhythmia disease; and 153 targets of drug components and disease were collected; the topological analysis was carried out by 2 times of the average degree(14.78) to obtain 4 key core targets:NR3C1,NOS3,ESR1,PTGS2; GO biological process and KEGG enrichment analysis showed that treatment of arrhythmia in this way mainly involved 375 biological processes and 34 KEGG pathways.The biological processes mainly involved cell plasma membrane,plasma membrane part,ion binding,intracellular signal cascade,response to organic matter,regulation of cell proliferation and other biological processes; its KEGG pathway was most likely to interact through neuroactive ligand-receptor interactions,followed by calcium signaling pathway and an adrenaline signaling pathway of cardiomyocytes.Conclusion:Danggui Liuhuang Decoction has the characteristics of multiple components,multiple targets and multiple pathways in the treatment of patients with arrhythmia.Its mechanism is relatively complex,and the therapeutic effect may be achieved mainly through the interaction of nerve active ligand-receptor. |
