      目的:研究扶正消积胶囊在体外对人胃癌细胞BGC-823增殖抑制及凋亡的影响。方法:MTT实验检测扶正消积胶囊对BGC-823细胞增殖的影响;流式细胞仪检测其对BGC-823细胞凋亡的影响;Real Time-PCR、Western blotting实验检测其凋亡相关基因表达。结果:与空白组比较,BGC-823细胞的增殖能被扶正消积胶囊显著抑制(给药24 h),此抑制率随给药剂量的增加而上升,在剂量为5 mg/ mL时,其抑制率达86.71%,且这种抑制呈剂量依赖关系。Annexin V/PI双染法联合流式细胞仪结果显示扶正消积胶囊能诱导BGC-823细胞凋亡,BGC-823细胞早期凋亡率随给药剂量的增加,在给药剂量分别为2.5 mg/mL、5 mg/mL时,BGC-823细胞早期凋亡率分别达到15.76%、38.25%;Western blotting、Real Time-PCR结果表明,Procaspase-3、9蛋白酶表达随剂量的增加而明显减小,Bcl-2、Bcl-xl、Survivin 3种基因表达随给药剂量的增加而下降,Bax基因表达则相反。结论:扶正消积胶囊能通过激活caspase级联反应抑制胃癌细胞增殖并诱导其凋亡。
English Summary:
      To investigate the influence of Fuzheng Xiaoji Capsule on the proliferation inhibition of human gastric cancer cell BGC-823 and its apoptosis.Methods:Effects of Fuzheng Xiaoji Capsule on proliferation of BGC-823 cells were investigated with MTT assay.Flow cytometry was used to detect the cell apoptosis while the effect of apoptosis related gene of Fuzheng Xiaoji capsule on BGC-823 cell was measured by real time-PCR and Western blotting.Results:MTT assay showed that the BGC-823 cells were inhibited by Fuzheng Xiaoji Capsule compared with control group,after treated on 24 h.The inhibition rate increases with the increase of dose,which is dose dependent.At the dose of 5 mg/mL,the inhibition rate reached 86.71%(P<0.05).Flow cytometry demonstrated that Fuzheng Xiaoji Capsule could induce the apoptosis of BGC-823 cells.The early apoptosis rate of BGC-823 cells increased with the dose of the drug.At the dose of 2.5 mg/mL and 5 mg/mL,respectively,the early apoptosis of BGC-823 cells reached 15.76% and 38.25% respectively.By the results of Western blotting and real time-PCR,the expression of caspase-3、caspase-9 was significantly lower as the dose increased.Also,it could reduce the activity of Bcl-2,Bcl-xl,Survivin and increase the activity of Bax.Conclusion:Fuzheng Xiaoji Capsule could inhibit the proliferation of BGC-823 cells and induce their apoptosis.The possible mechanism was through activation of caspase cascade reaction.
