Abstract:To explore the effects and the underlying mechanism of Buyang Huanwu Decoction(BYHWD) antioxidant function in spinal cord injury rats.Methods:A total of 24 male rats were randomly divided into the following 4 groups:a control group,a spinal cord injury model group(SCI group),a low-dose of BYHWD group(BL group),a high-dose of BYHWD group(BH group),with 6 rats in each group.The rat model was established by swimming exhaustion combined with the transection of rubrospinal tract(RST).The rats in control group and SCI group were given normal saline by gavage once,and the other groups were given BYHWD by gavage once.The recovery of motor function was observed by spontaneous vertical exploration test.MDA and SOD in serum was detected by ELISA.laser scanning confocal microscopy was adapted to detect the expression of CREB and BDNF.Results:Compared with the SCI group,the utilization rates of both forelimbs and the right forelimb of the BH group were higher than those of the SCI group and the BL group(P<0.05 or P<0.01).Compared with the SCI group,ELISA showed that the expression of MDA were decreased in the treatment group,while the expression of SOD was increased,in which the BH group were most significant(P<0.05 or P<0.01);Compared with the SCI group,Laser scanning confocal microscopy showed that the expression of CREB and BDNF were increased,in which the BH group were most significant(P<0.05 or P<0.01).Conclusion:BYHWD could improve the antioxidant function through the CREB/BDNF pathway in spinal cord injury rats.
