目的:观察桃红四物汤对心肌梗死后心室重构的作用以及对血管紧张素受体1(AT1)表达的影响。方法:通过结扎大鼠冠状动脉构建心肌梗死后心室重构模型。将大鼠随机分为假手术组、模型组、卡托普利组、桃红四物汤低、中、高浓度组,每组12只,每组大鼠连续给药12周。治疗结束后采用多普勒超声心动图评价大鼠心功能,计算心脏质量/体质量比率,采用苏木精-伊红和Masson染色观察心肌组织的形态变化和胶原沉积水平,并通过免疫组织化学法观察胶原蛋白I和胶原蛋白Ⅲ的表达,Western Blotting检测心肌组织中AT1的表达水平。结果:冠脉结扎后大鼠出现明显的心脏结构改变和功能障碍。与假手术组比较,模型组大鼠出现心肌组织损伤,间质纤维化,而左室舒张末期内径(LVEDD)、左室收缩末期内径(LVESD)显著升高(P<0.01),而射血分数(EF)、短轴收缩率(FS)显著降低(P<0.01),心脏质量指数显著升高(P<0.01),同时胶原水平升高(P<0.01),胶原蛋白Ⅰ/Ⅲ表达比值显著增加(P<0.01),心肌组织中AT1表达水平显著上调(P<0.01)。与模型组比较,桃红四物汤各浓度组与卡托普利组均可显著改善心肌组织的病理变化,提高EF和FS,降低LVEDD和LVESD,降低心脏质量指数,抑制心肌组织中过度胶原形成和胶原蛋白Ⅰ/Ⅲ表达比值,同时显著降低心肌组织中AT1表达水平(P<0.05或P<0.01)。结论:桃红四物汤可以显著降低心肌梗死后心肌纤维化,改善心室重构,其机制可能与其对AT1的抑制作用有关。 |

To observe the therapeutic effects of Taohong Siwu Decoction on ventricular remodeling and angiotensin type I receptor(AT1) in rats with myocardial infarction.Methods:Model of ventricular remodeling after myocardial infarction was established by ligating the anterior descending coronary artery in wistar rats.After the operation, the rats were randomly divided into a sham operation group, a model group, a Taohong Siwu Decoction low, medium, and high dose treatment group, and a captopril treatment group, with 12 rats in each group.Rats in each group were given continuous administration for 12 weeks.At the end of the treatment, doppler echocardiography was used to evaluate the heart function of the rats, the heart mass/body mass ratio was calculated, and the morphological changes and collagen deposition levels of myocardial tissue were observed by HE and Masson staining.Immunohistochemistry was used to observe the expression of collagen I and collagen Ⅲ.The expression level of AT1 in myocardial tissue were detected by Western Blotting.Results:After coronary artery ligation, rats showed obvious changes in cardiac structure and dysfunction.Compared with the sham operation group, rats in the model group had myocardial tissue damage and interstitial fibrosis.In addition, the left ventricular end systolic diameter(LVESD) and left ventricular end diastolic diameter(LVEDD) in model group were significantly increased(P<0.01), while the ejection fraction(EF) and short-axis contraction rate(FS) was significantly reduced(P<0.01).Heart mass index increased significantly(P<0.01), while collagen content increased(P<0.01).Collagen Ⅰ/Ⅲ expression ratio increased significantly(P<0.01), and AT1 expression level in myocardial tissue was significantly increased(P<0.01).Compared with the model group, each concentration group of Taohong Siwu Decoction and the captopril group can significantly improve the pathological changes of myocardial tissue, increase EF and FS, reduce LVEDD and LVESD, reduce heart weight index, and inhibit excessive collagen in myocardial tissue formation and collagen Ⅰ/Ⅲ expression ratio, while significantly reducing the expression level of AT1 in myocardial tissue(P<0.05 or P<0.01).Conclusion:Taohong Siwu Decoction can significantly reduce myocardial fibrosis after myocardial infarction and improve ventricular remodeling.The mechanism may be related to the inhibitory effect on AT1. |
