目的:运用网络药理学的分析方法探讨淫羊藿-巴戟天治疗勃起功能障碍的潜在机制。方法:从中药系统药理学数据库与分析平台(TCMSP)、生物信息学分析工具BATMAN-TCM数据库筛选出淫羊藿、巴戟天的有效活性成分及作用靶点,运用GeneCards、OMIM、NCBI数据库获取勃起功能障碍的疾病靶点,韦恩图筛选药对、疾病的共同的靶点。采用String数据库进行蛋白质-蛋白质相互作用(PPI)网络构建,Cystoscap 372进行拓扑分析,构建药对中药-活性成分-疾病-靶点交集网络,运用R363软件对共同靶点进行基因本体(GO)富集分析和京都基因和基因组百科全书(KEGG)富集分析,筛选出潜在信号通路并分析其作用机制。结果:1)巴戟天、淫羊藿筛选出203个靶点。2)勃起功能障碍疾病筛选出1 004个靶点。3)淫羊藿-巴戟天药对与勃起功能障碍的交集靶点共75个。4)通过PPI网络筛选出IL6、VEGFA、CASP3等30个关键靶点。5)GO分析的生物过程主要富集在对脂多糖的反应等过程;细胞组分主要富集在膜筏、膜微区等部位;分子功能主要富集在肾上腺素能受体活性等。6)KEGG分析共富集到PI3K-AKT信号通路等17条信号通路。结论:淫羊藿-巴戟天药对治疗勃起功能障碍,主要通过影响PI3K-AKT等信号通路发挥作用,同时可能与降低精氨酸酶、乙酰胆碱酯酶和腺苷脱氨酶的活性,上调NO水平,抑制PDE-5活性及影响细胞信号转导等机制相关。 |

To study the target and signal pathway of Epimedium-Morinda in the treatment of erectile dysfunction,and to explore the potential mechanism of Epimedium-Morinda in the treatment of erectile dysfunctionMethods:The active components and targets of epimedium-Morinda were screened out from TCMSP and BATMAN-TCM database,the disease targets of erectile dysfunction were obtained by Genecards,OMIM and NCBI gene human database,and the common targets of drugs and diseases were screened by venntuThe PPI network was constructed by using String database,and the topological analysis was carried out by using Cystoscap 372The intersection network of traditional Chinese medicine,active ingredient,disease and target was constructedThe common target was analyzed by using R363 software for GO analysis and KEGG pathway enrichment analysis,and the potential signal pathway was screened and its mechanism was analyzedResults:1)Morinda has 20 chemical constituents and 43 targetsEpimedium has 23 chemical constituents and 202 targetsAfter merging and deleting,203 targets are obtainedEffective ingredients include quercetin,kaempferol,sitosterol,luteolin,etc2)1004 targets were selected for erectile dysfunction3)There were 75 intersection targets of Epimedium-Morinda and erectile dysfunction4)A total of 30 key targets such as IL6,VEGFA and CASP3 were selected through PPI network5)The biological process of GO analysis was mainly concentrated in the reaction to lipopolysaccharide,the reaction to bacterial source molecules,the vascular reaction in the circulatory system,etc; the cell components were mainly concentrated in the membrane raft,membrane micro area,membrane area,presynaptic membrane and posterior membrane,etc; the molecular function was mainly concentrated in the activity of adrenergic receptor,G protein coupled amine receptor,cytokine activity,nuclear receptor,transcription factor activity,etc6)KEGG analysis enriched 17 signaling pathways,including PI3K AKT signaling pathway,neuroactive ligand receptor interaction,calcium signaling pathway,NF kappa B signaling pathway,cytokine cytokine receptor interaction,HIF-1 signaling pathway,p53 signaling pathway,cGMP-PKG signaling pathwayConclusion:Epimedium-Morinda plays an important role in the treatment of erectile dysfunction by affecting PI3K AKT and other signaling pathways,and may be related to the mechanisms of reducing the activities of arginase,acetylcholinesterase and adenosine deaminase,increasing the level of NO,inhibiting the activity of PDE-5 and affecting the cell signaling. |
