      目的:建立芎附滴丸(XFDP)的质量标准,并对其进行抗偏头痛的药效学研究。方法:采用高效液相色谱法(HPLC)同时测定XFDP中8种主要成分的含量。评价XFDP对偏头痛大鼠5-羟色胺(5-HT)、β-内啡肽(β-EP)、降钙素基因相关肽(CGRP)、内皮素-1(ET-1)、基质金属蛋白酶-9(MMP-9)、核因子κB(NF-κB)、一氧化氮合酶(NOS)含量,及Calca、c-fos mRNA表达的影响。结果:建立的HPLC法线性关系、精密度、重复性、稳定性良好,各成分的平均加样回收率和相应的RSD分别为阿魏酸100.68(2.76%)、洋川芎内酯I 98.71(2.88%)、洋川芎内酯A 101.66(2.17%)、丁基苯酞100.15(2.60%)、Z-藁本内酯99.50(2.88%)、Z-丁烯基苯酞102.47(2.22%)、香附烯酮100.11(2.91%)、α-香附酮100.66(3.03%)。偏头痛造模后大鼠5-HT、β-EP含量显著降低(P<0.01,P<0.01),CGRP、ET-1、MMP-9、核因子κB、NOS含量显著升高(P<0.05,P<0.05,P<0.01,P<0.01,P<0.01),Calca、c-fos mRNA的表达显著升高(P<0.05,P<0.01)。XFDP可升高模型大鼠5-HT、β-EP含量,降低CGRP、ET-1、MMP-9、核因子κB、NOS含量,降低Calca、c-fos mRNA的表达,而发挥治疗偏头痛的作用。结论:本方法简单、快速、准确,可用于XFDP的质量控制;XFDP通过调节多种神经递质和血管活性物质的表达发挥治疗偏头痛的作用。
English Summary:
      To establish quality standards and evaluate the anti-migraine effect of Xiongfu dripping pills.Methods:A HPLC method was used for simultaneous determination of 8 main components in Xiongfu dripping pills.The effects of Xiongfu dripping pills on the contents of 5-HT,β-EP,CGRP,ET-1,MMP-9,NF-κB,NOS and the mRNA expression of Calca and c-fos in migraine rats were evaluated.Results:The HPLC-DAD method determined the content of ferulic acid,senkyunolide I,senkyunolide A,3-butylphthalide,Z-ligustilide,Z-butylidenephthalide,cyperotundone and α-cyperone.The precision,repeatability and stability were favorable.The average recoveries and corresponding RSD values were 100.68(2.76%)、98.71(2.88%)、101.66(2.17%)、100.15(2.60%)、99.50(2.88%)、102.47(2.22%)、100.11(2.91%)、100.66(3.03%),respectively.Compared to normal rats,the levels of 5-HT and β-EP in migraine rats decreased significantly(P<0.01,P<0.01),CGRP,ET-1,MMP-9,NF-κB and NOS increased significantly(P<0.05,P<0.05,P<0.01,P<0.01,P<0.01),and the mRNA expression of Calca and c-fos also increased significantly(P<0.05,P<0.01).Xiongfu dripping pills significantly improved 5-HT and β-EP,reduced CGRP,ET-1,MMP-9,NF-κB,NOS and reduced the mRNA expression of Calca and c-fos to treat migraine.Conclusion:The HPLC method is simple,fast,accurate,and it can be used for the quality control of Xiongfu dripping pills.Xiongfu dripping pills treat migraine by regulating various neurotransmitters and vasoactive substances.
