      目的:基于网络药理学探讨四妙散治疗骨性关节炎(OA)的作用机制。方法:从中药系统药理学数据库与分析平台(TCMSP)中检索四妙散中黄柏、苍术、薏苡仁、牛膝的活性成分,以口服生物利用度(OB)≥30%,类药性(DL)≥0.18为条件进行筛选,通过Uniprot数据库进行靶点预测;通过TTD、Drugbank、CTD、Gencards、OMIM、PharmGKB、GAD、KEGG数据库获取OA的相关靶标,并进一步对药物和疾病靶点相映射得到四妙散治疗OA的潜在靶点,运用Cytoscape和STRING软件构建“中药-成分-靶点”与蛋白质-蛋白质相互作用(PPI)网络图,筛选可能发挥治疗作用的重要成分及靶点,并对相关靶点进行基因本体(GO)富集分析和京都基因和基因组百科全书(KEGG)通路富集分析获知其潜在的作用机制。结果:从四妙散中共筛选出活性成分67个,包括槲皮素、豆甾醇、β-谷甾醇、汉黄芩素、非洲防己碱等,与骨关节炎相关的作用靶点524个,包括TNF、IL6、MAPK1、PTGS2、MAPK8等。四妙散主要通过调节TNF信号通路、HIF-1信号通路、NOD样受体、Toll样受体信号通路等发挥治疗骨关节炎的作用。结论:本研究运用网络药理学预测了四妙散治疗OA可能的活性成分、关键靶点及作用通路,体现了中药治疗疾病具有“多成分、多靶点、多途径”的特点,为其药效物质基础和作用机制研究提供了理论基础。
English Summary:
      To explore the mechanism of Simiao Powder in the treatment of osteoarthritis based on network pharmacology.Methods:The active components of Phellodendron Phellodendri,Atractylodes,Coix seed and Achyranthes bidentata in Simiao Powder were retrieved from TCMSP database,screened under the condition of OB≥30%,DL≥0.18,and the target was predicted by Uniprot database.The related targets of osteoarthritis were obtained through the databases of TTD,Drugbank,CTD,Gencards,OMIM,PharmGKB,GAD and KEGG,and the important targets of Simiao Powder in the treatment of osteoarthritis were further mapped by drugs and disease targets.The PPI network of “traditional Chinese medicine-active ingredient-important target” and protein interaction was constructed with Cytoscape and STRING,and the main treating components and key targets were screened.The potential action mechanism of the targets was obtained by GO functional enrichment and KEGG pathway enrichment analysis.Results:A total of 67 active components were screened from Simiao Powder,including quercetin,stigmasterol,β-sitosterol,wogonin,Tetrandrine,etc,and 524 targets related to osteoarthritis,including TNF,IL6,MAPK1,PTGS2,MAPK8 and so on.Simiao Powder mainly played a role in the treatment of osteoarthritis by regulating signal pathways of TNF,HIF-1,NOD-like receptor,Toll-like receptor.Conclusion:In this study,the possible active components,key targets and pathways of Simiao Powder in the treatment of osteoarthritis were predicted by the method of network pharmacology,which reflects the characteristics of “multi-components,multi-targets and multi-pathways” in the treatment of traditional Chinese medicine.It provides a theoretical foundation for the study of its pharmacodynamic material base and functional mechanism.
