目的:借助网络药理学平台技术探究复健片治疗缺血性脑卒中(IS)的作用机制。方法:利用中药系统药理学与分析平台(TCMSP)、中医药综合数据库(TCMID)、Batman-TCM数据库筛选出复健片中何首乌、桑寄生、决明子、淫羊藿、海马的活性成分及其靶点;利用Uniprot数据库对复健片中靶点信息进行校对;运用GeneCards数据库获取IS的疾病靶点基因;通过Cytoscape 3.7.2软件及STRING 11.0数据库构建蛋白质-蛋白质相互作用(PPI)网络并进行生物富集分析。结果:获取复健片治疗IS的共45个活性成分和144个靶点,关键靶点涉及IL6、IL1β、TNF、VEGFA、NOS3等;IS相关靶点1 485个;获得300条GO生物过程和84条相关信号通路,涉及PI3K-AKT信号通路、TNF信号通路、cGMP-PKG信号通路、cAMP信号通路、核因子κB信号通路等。结论:复健片是通过多成分、多靶点、多通路相互作用的机制来达到治疗IS的目的,通过本研究为复健片面向临床进一步推广应用提供理论支持,并为未来研究提供思路。 |

To investigate the mechanism of Fujian Tablets in the treatment of ischemic stroke(IS) with the help of Internet pharmacology platform technology.Methods:TCMSP,TCMID and Batmann-TCM databases were used to screen the active components and their targets of Polygonum multiflorum,sangjis-heng,Cassia obtusifolia,epimedium and hippocampus in Fujian Tablets;the target information in Fujian T-ablets was proofread by UniProt database; the disease target genes of IS were obtained by GeneCard-s database; Cytoscape 3.7.2 software and STRING 11.0 database were used to construct PPI network and conduct bio-enrichment analysis.Results:A total of 45 active components and 144 targets were obtained,including IL6,IL1 β,TNF,VEGFA,NOS3,etc.; 1485 IS related targets; 300 GO biological processes and 84 related signal pathways were obtained,involving PI3K-AKT signaling pathway,TNF signaling pathway,cGMP-PKG signaling pathway,cAMP signaling pathway,NF-κB signaling pathway,etc.Conclusion:Fujian Tablet through the mechanism of multi-component,multi-target,multi-channel interaction can achieve the purpose of treating IS.This research provides theoretical support for the further clinical application of Fujian tablets,and provides ideas for future experimental research. |
