      目的:研究健脾止动汤对多发性抽动症(TS)模型大鼠外周血、结肠、皮质、纹状体细胞因子表达的影响。方法:采用亚氨基二丙腈(IDPN)腹腔注射法建立TS动物模型,将模型大鼠随机分为空白对照组、模型组、泰必利组、健脾止动汤组。模型组和空白对照组给予0.9%生理盐水灌胃,其余2组分别给予等体积的泰必利混悬液和健脾止动汤颗粒溶液,连续给药6周。给药结束后,采用酶联免疫吸附试验法(ELISA)分别检测大鼠外周血白细胞介素-1(IL-1)、白细胞介素-2(IL-2)、白细胞介素-4(IL-4)、白细胞介素-6(IL-6)、白细胞介素-10(IL-10)、γ干扰素(IFN-γ)、肿瘤坏死因子-α(TNF-α)及结肠、皮质、纹状体IFN-γ与TNF-α表达水平。结果:1)健脾止动汤组IL-1、IL-2、IL-6、IFN-γ、TNF-α表达显著低于模型组(P<0.05)和泰必利组(P<0.05);2)健脾止动汤组IFN-γ、TNF-α表达较模型组(P<0.05)和泰必利组(P<0.05)显著下降;3)健脾止动汤组IFN-γ、TNF-α表达较模型组(P<0.05)和泰必利组(P<0.05)显著下降,泰必利组IFN-γ、TNF-α表达较模型组无显著变化(P>0.05)。结论:IDPN诱导TS模型存在免疫功能紊乱,健脾止动汤可抑制外周和中枢组织多种炎症介质释放,其抑炎作用可能对控制TS发作具有一定辅助调节作用。
English Summary:
      To study the effect of Jianpi Zhidong Decoction on the expression of cytokines in peripheral blood,colon,cortex and striatum of Tourette Syndrome (TS) model rats.Methods:The TS animal model was established by intraperitoneal injection of iminodipropionitrile (IDPN) and the rates was randomly divided into a blank control group,a model group,a Tiapride group,and a Jianpi Zhidong Decoction group.The model group and the blank control group were given 0.9% normal saline by gavage.The other 2 groups were given equal volumes of Tiapride suspension and Jianpi Zhidongtang Granule solution for 6 weeks.After the administration,enzyme-linked immunosorbent assay (ELISA) was used to detect IL-1,IL-2,IL-4,IL-6,IL-10,INF-γ,TNF-α and colon,The expression levels of INF-γ and TNF-α in the cortex and striatum.Results:1) The expressions of IL-1,IL-2,IL-6,IFN-γ,and TNF-α in the Jianpi Zhidong Decoction group were significantly lower than those in the model group (P<0.05) and Tiapride group (P<0.05); 2 ) The expression of INF-γ and TNF-α in Jianpi Zhidong Decoction group was significantly lower than that of model group (P<0.05) and Tiapride group (P<0.05); 3) INF-γ and TNF-The expression of α was significantly lower than that of the model group (P<0.05) and the Tiapride group (P<0.05).The INF-γ and TNF-α expressions of the Tiapride group had no significant changes compared with the model group (P>0.05).Conclusion:IDPN-induced TS model has immune dysfunction.Jianpi Zhidong Decoction can inhibit the release of various pro-inflammatory factors in peripheral and central tissues,and its anti-inflammatory effect may have a certain auxiliary regulatory effect in controlling the onset of TS.
